Nerve lesion resulted from peripheral nerve injury is commonly encountered clinical trauma, so autotransplantation of nerve became main treatment. But because regeneration of nerve isn' t perfect and recovery of nerve function was effected, especially because the donor resource of the autotransplantation of nerve is limited and autotrans-plantation of nerve can cause complicating disease such as neuralgia-numbness and scar in donor sites, domestic and international scientists have performed many researches on allograft of nerve to find a ideal graft to replace autotransplantation. In spite of a lot of new achieve-ments, immunologic rejection is still obstacle so far and there isnl breakthrough yet. The fact that the proliferation of connective tissue is faster than thatof nerve tissue may be the main problem. So in this ex-periment, base fibroblast growth factor ( bFGF) compound degradative membrane, which can improve the regenerative ability of nerve, delay the appearance of immunologic rejection and suppress the proliferation of connective tissue , was used to do a comprehensive function re-search , to improve the effect of regeneration of nerve.Material1. Animal:30 healthy big - ear white rabbits, weight 2500 -3000g, including male and female, bought from animal breeding cen-ter of China Medical University.2. Main reagents: bFGF compound degradative membrane: bFGF bought from Zhuhai bio - product corporation, prepared together with Shenyang Pharmacy University.3. Main Instrument:(1) Electromyogram instrument: from Denmark.(2) Image pattern analysis instrument; from China Shichuan computer institution.(3) Electron microscope: from Japan.(4) Optical microscope:from Japan.Method1. Left right median nerves of 30 rabbits, total 60 nerves, were excised by 2.5 cm. Animals were divided into 4 groups( A B C D) : Group A and group B had 15 rabbits and 30median nerves were in-jured. All the excised nerves were immersed in receptor blood plasma for 20 minutes, then were stored in - 196?C liquid nitrogen for 21 days, after the excised nerves recovered to room temperature, heter -median -nerve suture were operated at room temperature, the differ-ence between group A and group B is followed, left median nerves transplantation sites in group A were packed with bFGF compound slow - released degradative membrane as experimental group, right median nerves transplantation sites in group B weren' t packed as con-trol; Group C and group D had 15 rabbits and 30 median nerves were injured, all the excised nerves were stored in -196?C liquid nitrogen for 21 days, after the excised nerves recovered to room temperature, heter ?median ?nerve suture were operated at room temperature, thedifference between group C and group D is followed, left median nerves transplantation sites in group C were packed with bFGF com-pound slow ?released degradative membrane as experimental group, right median nerves transplantation sites in group D weren' t packed as control. In the different time after operation, we can observe following faction; Macroscopic observation; Microscopic observation; Luzex - F image pattern analysis instrument determination; Quantitative determi-nation of peripheral joint of far near nerve suture segment; Observa-tion of blood ?nerve barrier function recovery and Electro - physiolog-ical index examination.2. Statistical method;Perform t - test, analysis of variance by STATISTICA5. 0 soft-ware.Rrsults1. Visual inspection: we can see that: It appears more smooth in the A group and C group which is encapsulated by bFGF compound degradation membrane than B group and D group without encapsula-tion. And the fiber connected to peripheral tissue of A and C group is less than the B and D group, moreover, the spindle enlargement a-round the suture of proximal and distal nerve are obviously bigger than the A and C group, and adhesion to peripheral tissue are more severe than A and C groups.
