Propolis is often used in folk medicine as antiseptic and antibacterial. It possesses many biological activities:antibacterial,anti-inflammatory,regulating immunity,which has been well known by many researchers. The anti-inflammatory effects of propolis solution extracted by ethanol(EEP) and water(WSP) is studied in this thesis by means of modern pharmacological methods. While probing into anti-inflammatory effect and mechanism of action by means of advanced modern scientific research methods such as Enzyme-linked immunosorbent assay,fluoro-spectrophotometry,ultraviolet-spectrophototometry,immunohistochemistry and in situ hybridization at the level of cell and molecule. The results of the experiments show as following:1. EEP and WSP show the inhibiting function on the permeability of blood capillary of inflammatory tissue and the edemas which are induced by carrageenan and Freund's adjuvant complete. EEP and WSP have an obvious protective effect on the "twice-hit" animal model of acute lung injury in rats induced by oleic acid and lipopolysaccharide(LPS) from lung edema. They show excellent inhibition function on the chronic granuloma as well. The results of the experiments suggest that EEP and WSP have a good effect of anti-inflammation.2. EEP and WSP can decrease the contents of inflammatory mediators (PGE2 and total protein) and the level of lipid oxide products (MDA) in inflammation tissue. WSP can reduce the number of leukocyte and alleviate the inflammatory response. EEP can decrease the contents of cholesterol in adrenal glands,but it doesn't show significant influence on content of VitC. The inhibition effects on inflammation tissue of EEP may depend on the pituitary-adrenal system.3. EEP and WSP can inhibit the increase of NO,lysozyme contents and cytokines such as IL-6 in acute inflammatory tissue fluid. They show no significant influence on IL-2 and IFN- v contents. It suggests that they may action on B lymphocyte system.4. EEP and WSP decrease the expression of CD54 in the bronchial epithelium tissue and inhibite the activation of NF-kB p65 and expression of its mRNA in the bronchial epithelium tissue ofacute lung injured rat. They protecte the tissue from farther injuring by controlling the key step to inflammatory response.5. It seems that the EEP and WSP show the same effects on anti-inflammatory by different mechanism. WSP can alleviate the inflammatory response by decreasing the number of leukocyte. The inhibition effects on inflammation tissue of EEP may depend on the pituitary-adrenal system.6. Acute toxicity testing of EEP and WSP has been demonstrated that the maximal tolerance dose exceeds normal dose more than 400 times,which can be considered nontoxicity. There are no significant effects on immune organ in the course of giving drugs. And they can antagonize the body weight loss when creating the model.At the same time,the research direction in exploitation and development are discussed. The propolis has a great potential in healthy foods and medicine. The research results provide the science basis to propolis using as folk medicine and the pharmacology basis to using propolis as drugs.
