JingFang San (荆防散), be composed of Jingjie (荆芥) and Fangfeng (防风) , has effects of evapning wind-cold, and dispelling wind and arresting itching. In clinic,JingFang San or the prescriptions including JingFang San as principal component are used to treat allergic inflammation disease such as bronchitis, allergic rhinitis, and urticaria so on.Objective: In this study, the anti-inflammatory and anti-anaphylaxis effects of JingFang Sani, and partial mechanisms were investigated.Methods: The anti-inflammatory effects were studied by means of auricle edema induced by xylene in mice, the increasing of celiac capillary permeability induced by acetic acid in mice, paw edema induced by carrageenin in rats, Pleurisy induced by carrageenin in rats and the granulation caused by cotton-ball in mice, while the anti-anphlaxis effects were evaluated by using the passive cutaneous anaphylaxis (PCA) model in rats or mice and the delayted Hypersensitivity in mice. Finally, to investigate the mechanism of anti-inflammatory and anti-anphlaxis effects in two animal inflammation models carried out by the carrageenan-induced pleurisy in rats and ovabumin-induced asthma in mice.Results:1. JingFang San had anti-inflammatory action. JingFang San at 5, 2.5 and 1.25g/kg doses significantly inhibited auricle edema induced by xylene in mice, the increasing of celiac capillary permeability induced by acetic acid in mice, paw edema induced by carrageenin in rats. JingFang San at 5, 2.5 g/kg doses significantly reduced amount of ceukocyte and exudation protein in pleurisy model in rats. JingFang San at 5, 2.5 and 1.25g/kg doses significantly inhibited the granulation caused by cotton-ball in mice.2. JingFang San had anti-anphlaxis effect. JingFang San at 5, 2.5 and 1.25g/kg showed significant suppressive effect on the PCA reaction in rats or mice, and delayted hypersensitivity induced by DNCB in mice.3. Compared with the model group, JingFang San at 5g/kg dose significantly inhibited PGE2,LTB4 in pleurorrhea in pleurisy model rats and lung tissue homogenate in asthma model mice. JingFang San at 5g/kg dose significantly inhibited 5-LO expression in lung tissue in asthma model mice induced by ovabumin. It is suggested that the anti-inflammatory and anti-anphlaxis effects mechanism of JingFang San was related to decrease the metabolites of arachidonic acid.4. Compared with the model group, JingFang San at the doses of 5, 2.5 g/kg could decreased the NOS activity and the NO level in inflammation area in different grade, which showed significant suppressive effect at 5g/kg dose. It is suggested that the anti-inflammatory and anti- anphlaxis effects mechanism of JingFang San was related to interfere in NO/NOS pathway.Conclusions: The results showed that JingFang San has the effects of anti-inflammatory and anti- anphlaxis, which the mechanism be related to decrease the the level of PGE2 and LTB4, and interfere in NO/NOS pathway. It is the pharmacology basis for its efficiency and clinical use of treating allergic inflammation disease.
