Objective: To investigate the effects of Bu Yang Huan Wu Decoction (BYHWD) on inflammatory cellular factors and neural cell apoptosis in cerebroischemic and reperfused injury in gerbils.Methods: Cerebral ischemic reperfusion model in gerbils was used. Bilateral carotid arteries were li gated for 15 minutes, then reperfused for 48 hours. The contents of IL-1 β, IL-6 and TNF- α in blood serums as well as NO, amino acid, the activities of Na+, K+-ATPase , Ca2+-ATPase, Mg2+-ATPase and NOS in cerebral tissues were detected and compared in different groups. Neural cell apoptosis of hippocampal gyms slides was observed under electron microscope 120 hours after reperfusion.Result:1.In comparison with ischemia-reperfusion (IR) and standardized-comparison(SC) group, IL-1β, IL-6 and TNF- α were significantly decreased in BYHWD group (all p<0.01). Compared with sham-operation (SO) group, IL-1β, IL-6 and TNF- α in BYHWD and SC group were significantly increased (p<0.01).2. There was no apoptic cell in SO group. Among six gerbils, 5 gerbils could be found apoptic cells or apoptic bordies in IR group. There was only one gerbil that could be found apoptic cells in BYHWD group. According to X2 test, IR group's cell apoptosis was significantly increased(p<0.01). The incidence of neural cell apoptosis of hippocampal gyms in BYHWD group was more reduced than that in IR group (p<0.01).3.The activity of Na\ K+-ATPase in BYHWD group was increased compared with IR group (p<0.05), but significantly decreased compared with SO group(p<0.05).4.The content of NO in BYHWD group was increased compared with IR group (p<0.01), but significantly decreased compared with SO group(p<0.05). The activity of NOS was obviosly increased but had no significance(p>0.05).5.Glu's content was significantly reduced in BYHWD group compared with IR group(p<0.05) but had no significance compared with SO group (p>0.05).Conclusion:1.BYHWD can make IL-1 β , IL-6 and TNF- α increased in blood serrums in cerebroischemic and reperfused injury after 48 hours in gerbils. BYHWD has a protective effect on nervous injury in cerebral ischemia and reperfusion by reduction of inflammatory cellular factors.2.BYHWD can counteract neural cell apoptosis after cerebral ischemia in gerbils .Therefore, BYHWD can impruve injury in cerebral ischemia and reperfusion3.BYHWD is able to prevent the decrease of Na+, K+-ATPase activities, increase NO content and NOS activity, decrease Glu content.BYHWD can counteract neural cell apoptosis after cerebral ischemia in gerbils and its mechanism maybe related to the improvement of cerebral energy metabolism, regulation of NO synthesis and antagonism of toxic effect excitatory amino acid.
