The generation, development and metastasis of tumor, related with many factors, is a complex multiple -phases process. CyclinDl and Cyclin - dependent kinase4 ( CDK4) are closely linked to the occurrence of tumor, and now their overexpressions have been found in a variety of human tumors.CyclinD1, encoded by the gene on chromosome 11q13, is part of the complex molecular system regulating the Gl - S transition point of the cell cycle. Its overexpression, a common finding in human carcinomas , leads to the generation, development of tumor. CDK4 is encoded by the gene on chromosome 12q and play an impotant role in the regulation of cell cycle. By acting in conjuction with protein kinase CDK4, CyclinDl may phosphorylate retinoblastoma (Rb) at the end of the Gl phase. The Rb gene product, pRb, undergoes a series of phosphorylation and dephosphorylation events in the course of the cell cycle. During G0 and G1 pRb is present in hypophosphorylated or un-phosphorylated form that are able to block entry into S phase. Hyper-phosphorylation by certain CyclinDl - CDK4 complexes inactivates pRb, allowing cells to proceed through the cell cycle.In the past few years, a lot of study confirmed CyclinDl and CDK4 play important roles in the generation, development and prognosis of tumor. It may be significant to detect the expression of CyclinDl and CDK4 in non - small cell lung cancer (NSCLC) , in order to prevent and discover tumor early. In this research, we studied the expression of CyclinDl, CDK4 in lung adenocarcinoma ( AC) and lung squamous carcinoma ( SQC ) and discussed the relation between the expression of them and clinicopathologic factors of lung carcinoma via IHC.Material and Mathod1. MaterialSpecimen:Total of 77 cases, all from the department of thoracic surgery of the first clinical hospital of China Medical University, between the period of 1998 -2002, including 36 AC and 41 SQC, were diagnosed by the department of pathology. The group was consisted of 54 males and 23 females, whose age ranged 37 -78, average 61.2. The specimens of 10 lung benign tumor were taken as control team. The specimen, fixed in 4% Formaldehyde, was buried in paraffin. No distant metastasis was found in the group and no patient received chemotherapy or radioptherapy.Main instrument:Resection machine ( BRIGHT5040 made in English) , GRANSEmicrowave oven, OLYMPUS BH2 microscope, etc.Reagent;Two primary antibodies, used in this experiment, were both from Beijing Zhongshan Biotechnology, Ltd. so did the immunohistochem-istry reagent box. The concentration of two primary reagents was 1:50 respectively. DAB was provided by the laboratory of thoracic department.2. MathodS - P Immunohistochemistry:Section, removal of paraffin of section, inactivation, antigen repair, stain, restain, dehydration, cover.Evaluation of the specimens ;The parts that stained positive were observed to lie in the nucleus and ( or) cytoplasm of the cell. The positive stained part show brown. Ten randomly chosen visual fields were observed on each section under microscope. The proportion of positive stained cells was calculated a-mong 100 tumor cells. (-) indicating negitive, no cell was stained positive; ( + ) indicating weak positive, less than 20% cells were stained positive; ( ++ )indicating positive, more than 20% cells were stained positive. The part of weak positive was classified into negative group.Statistical analysis;X test was applied to analyze the results of experiment. P <0. 05 represent marked statistical significance, and P <0. 01 represent extraordinary statistical significance.ResultNo benign tumor cell was stained positive; the positive rates were 54. 5% ( CyclinD1) and 72. 7% ( CDK4) in lung adenocarcinoma and lung squamous carcinoma. There was statistically significant difference between benign tumor tissues and lung cancer (P <0. 01) and a positive correlation between CyclnDl expression and CDK4(P <0. 01) .By studying the relationship between the expression of CyclinDl and cli...
