Objective: To study the immunohistochemical change of vascular endothelial growth factor , nm23-Hl and nuclear factor-kB P65 in human brain glioma and human brain metastatic carcinoma and their roles in the biological behavior of the tumors.Material and methods: Protein expression of VEGF, nm23-Hl And NFRBP65 were examined by SABC immunohistochemical technique in 18 cases of human brain glioma ,12 cases of human brain metastatic carcinoma and 6 cases of normal brain tissue and the correlation between the expression and the biological behavior of the tumors was comprehensively analyzed.Results: The normal brain tissues just expressed nm23-Hl and didnot expresse VEGF and NFkBP65. But all tissues of the tumors expressed VEGF, nm23-Hl and NFkBP65 . The level of these three protein in recurrent gliomas and high-grade gliomas was considerably higher than that in low-grade gliomas and normal brain tissues(p<0.01).The level of nm23-Hl protein in normal brain tissues was higher than that in low-grade gliomas, but lowwer than that in high-grade gliomas. The level of these protein in human brain metastatic carcinomas was higher than that in low-grade gliomas, lowwer than that in recurrent gliomas. All these difference was significant(p<0.01).But the difference of the expression of these three protein in human brain metastatic carcinomas and that in high-grade gliomas wasnot significant(p > 0.05). VEGF, nm23-Hl and NFkBP65 protein were mainly distributed in tumor cells. Furthermore, VEGF protein could be distributed in endothelial cells of glioblastmas and anaplastic astrocytomas and nm23-Hl in normal brain cells. The expression of VEGF,nm23-Hl and NFkBP65 was significantly related with the degree of differentiation(p < 0.01) and recurrence(p< 0.01) of glioma and with the progression and invasion of human brain metastatic carcinoma.Conclusion: Expression of VEGF, nm23-Hl and NFkBP65 is associated with the maligant progression ,invasion and angiogenesis of human gliomas and human brain metastatic carcinomas,and it may play a critical role in the recurrence of gliomas. Nm23-Hl may suppress the progression and the dissemination of tumor cells in normal brain tissues and low-grade gliomas and promote the progression and the dissemination of the tumors in high-grade gliomas, recurrent gliomas and metastatic carcinomas.
