Objective To know preliminary genotype and allele frequency distribution of D1S549, D3S1754 and D12S375 in Chinese Han population in Qingdao area and to study the three STR loci for genetic application.Method ACD-blood specimens were collected from the unrelated individuals in Qingdao . The DNA samples were extracted with Chelex method and were amplified by polymerase chain reaction (PCR) technique. The PCR products were analyzed by poly-acrylamide gel electrophoresis(PAGE) and displayed using silver staining.Result Eight alleles were found at D1S549 locus, eight alleles at D3S1754 locus and five alleles at D12S375 locus and 22, 19 and 14 genotypes respectively. No deviation from Hardy-Weinberg equilibrium was observed in the three loci. The heterozygosity observed of the three loci were 0. 842 6, 0. 944 4 and 0. 781 8, and the heteozygosity expected of them were 0. 798 8,0. 708 7 and 0. 75 respectively. The exclusion probability was calculated as 0. 659 2 for D1S549, and 0. 560 5 for D3S1754 and 0. 586 4 for D12S375. The discriminating power of the three loci were 0. 914 3, 0. 886 1 and 0.886 1. The polymorphism information content of them were 0. 779 9, 0. 668 7 and 0. 729 6. The family studies on the three loci were conformed to the Mendelian inheritance model. Comparison of the allelic frequencies with Chengdu Han population , there is a difference statistical significantly at D1S549 locus and no difference at D3S1754 and D12S375 loci by chisqure test analysis.Conclusion This study reveal the structure of the three loci and the obtained data are beneficial to understanding the population genetics in Chinese Han population. All of the three loci have higher the chance of exclusion and the discriminating power and they will be useful markers for individual identification and paternity test and for genetics purposes.
