| Objective:Rheumatoid arthritis(RA) forms synovial proliferative lesion,afterwards causes chronic synovial inflammation and destruction of the articular cartilage,which can eventually lead to the progressive damage of the joints and functional disability. The significant characters of RA were blood vessel hyperplasia and inflammatory cells infiltration. The latter results in the lesions of synovium,cartilage and bone. For this reason, the study of the pathogenesis and treatment of synovial lessions of rheumatoid arthritis is an important issue urgent to be solved at present.Now there are many therapeutic drugs and methods ,such as ,immunosuppressive-agent,non-steroid-antiinflammation-drugs(NSAIDS),corticosteroids,gene and biology treatment etc.These treatments all demonstrate efficacy, but the clinical specificity,the reliability of long-term usage and expensive price of them still pose an important clinical puzzle.In addition,the set-up of ideal animal models of rheumatoid arthritis is also urgent to be settled.In the study progress of RA,Corticosteroids are always clinically applied widely as the most mighty anti-inflammation drug.It's not only the factor of utmost importance in the acute episode of RA,but also has good effect on single large joints of RA.The recently research thought it's anti-inflammatory function arose from below several aspects besides its immunosuppression function and direct anti-inflammatory function.①It inhibits the synthese of nitrogen monoxide (NO),and NO is a important medium of inflammation in the pathological process of RA.②It regulates the function and living of cells and pahapes could accelerate the apoptosis of synovial cells.③It could induce the syntheses of cellular factor of Th2, and inhibit the syntheses of Th1,regulating the equilibrium between Th1 and Th2. Diprospan is a kind of long-acting corticosteroids and includes betamethasone sodium phosphate and it's derivative betamethasone dipropionate, and betamethasone dipropionate is difficult to resolved as a slowly-released store 。So diprospan can be valid for a long time. Diprospan is in an advantageous position than the other corticosteroids in clinical.The trial was designed to study the effect and mechanism of intra-articular injection diprospan therapying the adjuvant arthritis of rabbits,employing the rabbits adjuvant arthritis models which is most similar to RA of human now and comparing effects with prednisolone acetate and saline intra-articular injection. Our study would lay a foundation of theory to directing diprospan therapying RA.Method:72 Japanese white rabbits ,weighing 2~3kg,were first sensitized by intradermal injection of ovalbumin 0.5ml(10mg/ml) emulsified in Freund's complete adjuvant 0.25 ml(back or hip).After one week rabbits were sensitized by intradermal injection of ovalbumin 0.5ml (10mg/ml) emulsified in Freund's incompletes adjuvant 0.25 ml.After two weeks rabbits were injected in both knee-joints with ovalbumin 0.5ml(10mg/ml).After one week the knee joint presented the typical adjuvant arthritis : swelling of joint,step-up of skin temperature,hyperemia of skin ,even erythema of skin in the severious cases.Acute adjuvant arthritis of both knee-joints of rabbits was induced.These rabbits were randomly assigned to three groups.There are 24 rabbits of every group .They are diprospan group(A),by both knee intra-articular injection diprospan 1 ml(1mg); prednisolone acetate group(B),by both knee intra-articular injection prednisolone acetate 1 ml(10 mg);control group(C),by both knee intra-articular injection saline 1 ml.Each group was taken 6 rabbits randomly,there were 18 totally.
