The metastasis of tumor is the peculiar behavior of biology for malignant tumor, which is the main reason of failure of treatment in clinic, and it affects directly on the prognosis and quality of life. The mechanism of metastasis of gastric carcinoma (GC) is still not very clear though many valuable studies have been reported. It is showed that metastasis of cancer is complex process, which is related to not only out of control in expression of oncogene and tumor suppress gene but also the functional defect of immune surveillance and the expression of adhensive molecule and changes of DNA plerosis gene and so on. We detected the expression of c-met, p27, CD62P, S-100 in gastric carcinoma and precancerous lesion using immunohistochemistry, and analyzed DNA ploidy to explore the correlation between DNA ploidy and polygene, at the same time, follow-up was done. The purpose of present study was to investigate the correlation between polygene variation and gastric carcinoma at the level of protein and gene.The results are as follows, l.The relationship between disease of chronic gastric mucosa lesion and the expression of c-met, p27, CD62P as well as S-100. The immunohistochemistry results showed that, there was increasing tendency with c-met protein positive expression during gastric mucosa malignant changes process from chronic superfical gastritis (CSG), chronic atrophy gastritis and intesting metaplasia (CAG+IM),dysplasia hyperlasia gastritis (DYS) to gastric carcinoma, p27, S-100 positive expression was decreasing in the pocess mentioned above. There was no significant difference with c-met, p27, and S-100 positive expression among CAG+IM, DYS and CSG groups (p>0.05). The expression of c-met in GC was significantly higher than that of CSG (p<0.01), but there was no difference with c-met expression between GC and the groups of CAG+IM and DYS (p>0.05). The expression of p2' in GC was obviously lower than that of CSG and CAG+IM (p<0.05), but there was no dramatic difference compared with DYS group (p>0.05). There was not significant difference with the expression of S-100 between groups mentioned above (p>0.05). The expression of CD62P in non-cancer was negative 2. The relationship between the expression of c-f\ -jmet, p , CD62P, S-100 and the biological behavior, prognosis of gastric carcinoma. The immunohistochemical result showed that the expression of c-met, CD62P and S-100 was correlated with the lymph nodemetastasis (p<0.05), and the expression of p27 was correlated closely with the lymph node metastasis (p<0.01). The expression of CD62P and p27 was correlated with the differentiation degree of GC (p<0.05), but the result of expression of c-met, S-100 is not related to it (p>0.05). The expression of p27, CD62P was correlated to pathological staging of GC (p<0.05), and the expression of c-met was significantly correlated to pathological staging of GC (p<0.01), but the expression of S-100 was not related to it (p>0.05). 3. The relationship among the expression of the c-met, p27, CD62P and S-100. There was positive correlation between the expression of c-met and CD62P (p<0.01). And there was negative correlation between the expression of c-met and p27(p<0.05). There waspositive correlation between the expression of p27 and S-100 (p<0.01), and there was negative correlation between the expression of p27 and CD62P(p<0.05). 4. The relationship between the type of the DNA ploidy and the expression of gene. The expression of CD62P in heteroploid groups was higher than that of diploid (p<0.01), and the expression of p27, S-100 in heteroploid was lower than that of diploid (p<0.05). 5. The relationship between the type of DNA ploidy and the biological behavior of gastric carcinoma. The type of DNA was closely correlated with the differentiation degree and the metastasis of lymph node (p<0.01 ),and it was correlated with the pathological staging (p<0.05). 6.
