Objectives Gastric carcinoma is commonly found in gastrointestinal cancer, it threatens the health of human. This study is to explore the relationship between heparanase(HPA) and nm23-Hl in gastric carcinoma , and judge whether they may be used as valuable markers in predicting postoperative metastasis and recurrence of gastric carcinoma.Methods Reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry (S-P method) were used to measure the expression of the HPA mRNA and nm23-H1 protein in the primary tumor tissue of 46 cases and the paracancer tissue of 39 cases, which were collected in QiLu hospital from Oct, 2003 to Mar 2004.Results The expression of HPA gene was positive in 26 cases of primary tumor tissue of gastric carcinoma with a positive rate of 56.5%, significantly higher than those in paracancer tissues of gastric carcinoma cases and in normal controls (P<0.01 ) . The HPA expression was not related with the size of tumor, envelope formation. The positive group showed a significantly differentiation in gross appearance, depth of invasion, histology and growth stale, lymph node metastasis, peritoneal dissemination and TNM stage, But no significant difference in tumor size. The positive expression rate of nm23-Hl protein ingastric carcinoma tissue is significantly lower than that in paracancer normal gastric tissue (P<0.05) .The low expression of nm23-Hl showed a significant differentiation in depth of invasion and TNM stage. The patients of HPA positive and nm23-Hl low expression have positive rate in peritoneal dissemination, lymph node metastasis.Conclusions (1) HPA shows high expression in gastric carcinoma tissue(2) The positive group shows a significant differentiation in gross appearance, depth of invasion , histology and growth stale , lymph node metastasis , peritoneal dissemination and TNM stage. HPA takes a key role in invasion and metastasis of gastric carcinoma cells.(3) Nm23-H1 shows lower expression in gastric carcinoma tissue than that of normal tissue.(4) Expression of HPA and/or nm23-Hl are related with metastasis and recurrence of gastric carcinoma. Combining the expression rate of HPA with the expression rate of nm23-Hl may predict gastric carcinoma postoperative metastasis and recurrence.(5) The positive rate of nm23-Hl without significant difference is found between HPA positive and negative group.
