Objective By detecting the expression of tumor suppressor gene PTEN, MAPK and cell cycle reglation factor cyclin Dl in lung cancer on the level of protein ,we investigate the relationship between these proteins and clinical factors,which may provide a clue to diagnosis and prognosis of lung cancer.Method All pathological slides of 62 cases, including 32 cases of clinical and following-up data,was reviewed before we started to perform immunohistochemical staining for PTEN,MAPK, and Cyclin Dl.The results were analyzed by Cox-Model and single factor Kaplan-Meier assay.Results (1) The positive rate of expression of PTEN in lung cancer was 75%. The expression level of PTEN was significantly correlated with the size of tumor and the lymph node metastasis and status of smokers. The expression level of PTEN was significantly correlated with the differentiation degree of NSCLC, (2) The positive rate of expression of MAPK in lung cancer were 33.87%. The expression level of MAPK was significantly correlated with the lymph node metastasis and the differentiation degree of NSCLC . (3) The positive rate of expression of Cyclin Dl in lung cancer were 66.13%. The expression level of Cyclin Dl was significantly correlated with smoking and the lymph node metastasis. (4) A significant negative relationship was observed between the expression of PTEN protein and the phosphorylation level of MAPK in Jung cancer .(5)Smoking, the differentiation , the expression of PTEN protein and TNM stage were highly hazardous facter in 11 facter of lung cancer of following-up case by Cox-Model analysis.(6) Cases of PTEN negative expression had a shorter survival time than those with PTEN positive expression by Kaplan-Meier assay and Wilcoxon method analysis.Conclusions Positive expression of PTEN, MAPK , CyclinDl protein was frequently event in lung cancer. There was a signaling transduction pathway by PTEN protein modulating the phosphorylation level of MAPK in development and progression of lung cancer. The altered expression of PTEN was significantly correlated with prognosis.
