Objective: To explore the function of interleukin-1β, CD11a and CD11b in leukostasis and tissue infiltration by leukemic cells, thereby to provide an evidence for clinical treatment and prevention. Methods: ELISA( enzyme linked immunobsorbent assay ) was used to investigate the contents of IL-1β in serum of 15 healthy people and 51 newly diagnosed or relapsed patients with acute myeloid leukemia (AML), who did not receive induction chemotherapies. Bone marrow mononuclear cells which contained more than 80% blast cells were obtained from 40 patients. The expression of adhesive molecules CD11a and CD11b on the bone marrow mononuclear cells were evaluated using immunocytochemstry. Results: The content of IL-1β in AML serum [82.82±72.30 (pg/ml) ] was higher than that of in the control group[27.44±13.67 (pg/ml) ], which differ significantly. There was a higher level for IL-1β in infiltrated group[l 14.22±87.47 (pg/ml) ] than in the other group[58.99±47.27 (pg/ml)], and there was a significant difference. IL-1β and peripheral blood cells were in linear correlation (r=0.796 ,p<0.01).There was no significant difference among the different FAB classes about the expression of CD11la, but the expressions of CD11b on M4-5 were higher than those of on the M1-2 and M3. In clinical characters ,patients whose CD11la were higher than 30% had higher peripheral blood white cells and infiltration than ones with lower CDl la .The positivity of CD11b had relationship with age, platelet and infiltration. IL-1β, CD11a and CD11b evidently correlated mutually (p<0.05). Conclusion: Intern including CDl la and CD11b play an important role in the adhesion of leukemic cells, leukemic cells can produce interleukin-1β , which can stimulate the expression of CD11a and CD11b. These suggest that the application of IL-1Ra (IL-1 receptor antagonist) and adhesion receptor antagonists may prevent and treat the occurrence of infiltration .
