Objective Detecting the expression of tumor suppressor gene p16 and Rbin cervical cancer(CC) and in cancer in situ (CIS), and analyze their relationship with expression of HPV16/18E7, to obtain further basic theoretical and experimental foundation for cervical cancer screening and its diagnosis.Methord (1)HPV16/18 infection was detected using PCR method. (2)Expressionof p16mRNA, RbmRNA and HPV16/18E7mRNA was assessed by reverse transcription polymerase chain reaction(RT-PCR). (3)Expression of P16, pRb and HPV16/18E7 protein were detected with immunochemistry technology (4)Data were analyzed with SPSS11.5 and Chinese statistic software.Results (1)The infection rate of HPV16/18 in CIS and CC was higher than normalcervical tissue (P<0. 05); statistically there was no difference between CIS and CC (P>0. 05) (2)Expression of HPV16/18E7mRNA and protein in CIS and CC is greater than in normal cervical tissue (P0. 05) (3)Expression of p16mRNA in CIS and CC is greater than in normal cervical tissue (P0. 05); expression of pRb in CIS and CC was lower than in cervical tissue. Statistically, no difference of Rb expression were observed among histological grade, age, size of tumor and mode metastasis.Conclusion (1)Overexpression of tumor suppressor p16 was occurred in CISand early invasive cervical cancer, it had strong correlation with expression of HR-HPV E7 mRNA. Overexpression of p16 may be a biomarker for cervical cancer screening and diagnosis. (2)Many factors involved in carcinogenesis, HR-HPV infection is the main causes. p16/CDK-cyclin/Rb plays an important role in the development of CIN and CC. The affection of HR-HPV to Rb gene is occurred in posttranscriptional level. And affect the expression of p16 indirectly. (3) Alteration of p16 and Rb gene in carcinogenesis of cervical cancer is a rare event.
