The Study On MAU,β2 MG Of Urine And MMPs Of Kidney In Earlier Period Diabetic Rats

OBJECTIVE To study the ALB,β2－MG and kinney's MMPs of earlier period diabetic rats. Methods 1 The construction of DM rats models :To select 32 rats weighted 160-220 gram (provided by Drug inspection and study institute of Ji Lin province ).Feeding without limitation of female and male.7 days for suitability . To divide them into two groups (1) DM group :17 rats , abdominal cavity injecting with STZ(bought from sigma company)diluted by sodium citrate under the dosage of 50mg/kg , examining the blood sugar three days later , considering the model succeeding if the blood sugar is more than 16mmol/l and urine sugar positive .(2)Control group : 15 rats ,abdominal cavity injecting comparative volume sodium citrate buffer . Feeding with the Con group after making model succeeding ,affording standard diet ,drinking freely. 2,acquiring specimen : DM group and Con group were all placed in metabolism cages ,fasting 24 hours ,drinking water freely .Collecting urine ,recording the volume ,divided into two tubes , 3ml per tube prepared for ALB ,β2－MG . diethyl ether anesthesia worded ,blood sampling 2ml retroorbitally, detecting blood sugar ,full auto biochemistry analyzer detecting blood CRE,BUN. Taking the right kidney ,peeling the surface fat ,dropping into liquid state nitrogen ,prepared for MMPs. RESULTS 1 The blood sugar level of two groups is significantly different (P<0.01),serum CRE,BUN levels of two groups is not different ;2 logarithmic UAE level of DM group is higher than the of Con group ,DM β2－MG level is a little higher than the of Con group ,but it has not statistic sense ;3 MMP-2 enzyme resolved volume of DM group is significantly lower than the of Con group and is negative correlated with logarithmic UAE ,the correlation coefficient r=-0.56949,(P<0.02). CONCLUSION 1 ALB of DM rats is higher than the of Con group in the earlier period (3 weeks ),ALB can be used as a sensitive index of monitoring the earlier stage diabetic nephropathy.2 The MMPs enzyme resolve volume decreased in the earlier stage of DM rats ,and is negative correlated with logarithmic UAE .ALB increased gradually with the decline of MMPs activity .MMPs decreased , EMC piled up ,primary filtration membrane structure is destroyed ,especially the change of array structure of IV type collagen ,these made the double layer structure electric charge barrier dysfunction ,then MAU increased .
