Objective : To determine frequencies distribution of peroxisome proliferator-activated receptor- y coactivator-1 (PGC-1) gene Gly482Ser polymorphism genotype and allele in Han and Korean population of Yanbian area in China,and its relationship with type 2 diabetes mellitus.Methods -. Polymerase chain reaction-restriction fragment length polymorphism was used to test a coding variant Gly482Ser of PGC-1 gene in 39 Han people and 36 Korean people with normal glucose tolerant(NGT) ,and 20 Han patients and 27 Korean patients with type 2 diabetes mellitus (DM) .Case-contrast analysis was used to study the association between Gly482Ser polymorphism and type 2 DM. Results: (1) The A allele frequencies of NGT subjects were 48.72% for Han, 43.06% for Korean, and no significant difference was found between this two groups ( x 2=0.483, P>0.05); (2) The A allele frequencies of type 2 DM patients were 47.50% for Han, 40.74% for Korean. Allele frequency distribution of each ethnic was compared between DM group and NGT group. There were no significant differences ( x2=0.016, P>0.05 for Han; x2=0.068, P>0.05 for Korean); (3) In Korean type 2 DM group, fasting plasma glucose (FPG) levels in GG group were lower than those in GA/AA genotype ( x 2=22.78, P < 0.05) .Conclusion : There was no difference in frequencies distribution of PGC-1 gene Gly482Ser polymorphism between Han and Korean population of Yanbian area in China. Their allele frequencies were similar to Japanese, however, there were significant differences when compared with European White and Pima Indian. The Gly482Ser polymorphism was not associated with type 2 DM in Han or Koreanpopulation of Yanbian area in China, but the mutated A allele was likely associated with FPG levels of Korean type 2 DM patients.
