Objective The 2.2.15 cell line was used as an experimental model in vitro to study the anti-HBV activity of Carboxymethytl Pachymaram ( CMP ) on the secretion of HBsAg, HBeAg in the cultured 2.2.15 cell line in order to provide support for its clinic use.Methods First, cellular toxicity test was performed. After the 2.2.15 cell line was planted 24 hours, CMP and Alcilovir (ACV) were added. The mean half toxicity concentrations (TC50) of CMP and ACV to the cell line was determined by the method of cyctopothic effect after 4 day's observation. Based on this, pharmaceutical anti-HBV experiment was performed. Concentrations of 20mg/ml,12mg/ml, 6mg/ml, 3mg/ml, 1.5mg/ml with CMP and concentrations of 2mg/ml, Img/ml, 0.5 mg/ml with Alcilovir (ACV) were added into the cell culturing medium respectively, and the super serum of the cell culturing medium was collected on the 4th and 8 day. ELISA method was used to assay its inhibitory rates of the secretion of HBsAg, HBeAg after of culturing12 days.Results Experiments showed that the TC50 of CMP is 13.6 mg/ml to 2.2.15 cell line and the concentration for 50% inhibition of the secretion (IC50) of HBsAg and HBeAg were 4.45 mg/ml and 5.61 mg/ml in the cultured cell line 2.2.15, and TI were 3.06 and 2.42. CMP shows stronger effect on anti-HBV than Alcilovir (ACV).Conclusion CMP has good inhibition effect on the secretion of HBsAg, HBeAg in the cultured cell line 2.2.15. The inhibitory rates of the secretion of HBsAg, HBeAg of CMP became higher as concentrations rise. The inhibitory effect appeared on the 4th and lasted until the 12th day, and the strongest inhibitory effect appeared on the 8th day.
