Objective: To study the inhibitory effect of CSB on duck hepatitis B virus (DHBV) in vivo and HBV in vitro,and provide an experimental basis for developing a new drug for the clinical treatment of patientis with hepatitis.Methods: (1) The efftct of CSB on HBV in vivo: First day old Guangxi brown spot ducklings infected with DHBV were used as the hepatitis B animal model. Positive ducklings were detected by PCR seventh days after the infection, and were randomly divided into five groups,each consisting of ten ducklings: Corydalis Saxicola Bunting (CSB) high-dose group, middle-dose group, low-dose group, Aciclovir positive group, control group. Every group was i.g. for fourteen days. The quantity of DHBV-DNA in serum was monitored by FQ-PCR, and ALT and AST activity in serum were detected by improvement Reitman-Frankel Method before treatment(T0), seventh days(T7) , fourteenth days(T14) after treatment,and three days after stopping treatment(P3). Liver histopathological examination by hematoxylin and eosin stain was done three days after stopping treatment. (2) The effect of CSB on HBV in vitro: The chicken embryo culture were used to observe the inhibition of CSB on Hepatitis B Virus by hemagglutination test in terms of hemagglutination titer, through the test to observe the pharmacodynamics of CSB extract. The tests showed that CSB extract could protect the liver damage that caused by carbon tetrachloride rachloride and acetaminophen. Meanwhile it could also have effect on immune function and Bile flow in rats; the median lethal dose was determined. Results: (1) The efftct of CSB on HBV in vivo:CSB could significantly decrease the serum DHBV-DNA titer (P<0.05). Three days after stopping treatment,DHBV-DNA returned significantly in the middle-dose group and low-dose group, while DHBV-DNA in the high-dose group did not return. CSB inhibitory effects were dose-dependent and time-dependent.Before and after treatment, OD value of ALT and AST in serum changed as well as DHBV-DNA titer. In addition, Liver histopathological examination confirmed that CSB had significant protective effects against the liver injury induced by DHBV.(2) The effect of CSB on HBV in vitro:Compared with the vires control group, hemagglutination titer of the CSB group decreased,the inhibition effects on HBV of CSB gradually attenuate with reduction of mass concentration, CSB group can prevent and protect of being infected by hepatitis B virus in chick embryo culture. The damage of hepatic tissue was significantly decreased by the large dose of herba CSB extract; it had effect on the liver damage induced by acetaminophen; it could improve immune function obviously; the median lethal dose was 298.5mg·kg -1.Conclusions: CSB can inhibit DHBV in vivo and HBV in vitro significantly, this study provided the scientific evidence for the further research that CSB might to be a drug to anti-HBV and cure hepatitis B effectively.
