Objective: To study the inhibitory effect of LYXC on duck hepatitis B virus (DHBV) in vivo and HBV in vitro,and provide an experimental basis for developing a new drug for the clinical treatment of patients with hepatitis.Methods: (1) 1-day old Guangxi brown spot ducklings infected with DHBV were used as the hepatitis B animal model. Positive ducklings were detected by PCR 13 days after the infection,and were randomly divided into five groups:Liuyuexue Compound (LYXC) high-dose group,middle-dose group, low-dose group, model group, Aciclovir control group.Every group had 10 ducklings, and LYXC was given, i.g.,for 14 days.The quantity of DHBV DNA in serum was monitored by FQ-PCR,DHBsAg and DHBeAg in serum were detected simultaneously by ELISA. ALT and AST in serum were detected before treatment(T0),7days(T7) and 14days(T14) after treatment, 3 days after stopping treatment. Liver histopathological examination was done 3 days after stopping treatment. (2) The effect of LYXC on HBV in vitro:The TC50 and TC0 of LYXC to the HepG2.2.15 cells transfected with HBV gene was determined by MTT assay. Adding LYXC to cultured HepG2.2.15 cells by two different drug adding methods: adding directly and serum pharmacology method. HepG2.2.15 cells were cultured in medium containing different non-cellulotoxic concentration and 10% serum of LYXC. After 72h and 144h of incubation, the culture medium was collected for determining the quantity of HBV-DNA by FQ-PCR and the levels of HBsAg, HBeAg by ELISA.Results: (1) The effect of LYXC on DHBV in vivo: LYXC could significantly decrease the serum DHBV DNA,DHBsAg and DHBeAg. LYXC in the high-dose group and middle-dose group could significantly inhibit DHBV-DNA replication in vivo (P<0.05 or P<0.01), and had apparent inhibitory effect on HBsAg and HBeAg (P<0.01).
