With the increasing of the new compounds in the world, the environment mutagens and carcinogens are threatening people more seriously. It has been a hot topic for the scholars at home and abroad to search for the effective anti-mutation and anti-tumor active components from the natural plants. Codonopsis lanceolata (Sieb. et Zucc.) Benth. et Hook. F. is from Codonopsis Wall, ex Roxb. of Campanulaceae. It was reported that the aqueous extract of Codonopsis lanceolata had significant antimutagenic effect and Codonopsis lanceolata polysaccharide had anti-tumor effect to sarcocarcinoma Si go- But study of antimutagenic effect of grease part of Codonopsis lanceolata were accordingly not reported. In order to search for and make sure its antimutagenic effective components, both the petroleum ether part and ethyl acetate part of the ethyl alcohol extracts of Codonopsis lanceolata were studied in this experiment. The experiment studied their pharmacological effect and chemical constituents.The petroleum ether, ethyl acetate and n-Butanol parts of the ethyl alcohol extracts of Codonopsis lanceolata were firstly used to do the Ames experiment. The petroleum ether part and the ethyl acetate part were proved to be effective. Then we isolated compounds from the petroleum ether part and the ethyl acetate part, and the compounds were also used to do the Ames test.The results showed: ( I ) In Ames test, the petroleum ether and ethyl acetate parts both had significant antimutagenic effects and decreased reverse mutation rate of the positive mutations of strains TA100. (II) 6 compounds were isolated from the petroleum ether and ethyl acetate parts by the methods of silica gel column chromatography. Among them, 4 compounds was crystals and their chemical structures were identified by the methods of the physiochemical properties, spectrography and comparing with calibrating compounds. They were shown below respectively: (1) TaraxeroK (2) a -SpinastenroK (3) Heptacosanoicacid. (4)Oleanolic acid. Other 2 compounds were oily and were determined by GC/MS. Their chemical structures were identified as: (5)Ethyl hexadecanoate. (6) 9-Octadecyne. Among them, (III) Compound (3) and (6) was isolated from Codonopsis lanceolata for the first time. (IV) The results of Ames anti-mutagenesis test showed that compounds dX (2X ?had anti-mutagenic effects.Above all, compounds (3) and (6) are isolated from Codonopsis lanceolata for the first time. In addition to Codonopsis lanceolata polysaccharide, compounds(1).(2).(4) also have antimutagenic effects. Other effective components remain to be studied in the future.
