| Objective To explore the pathogenisis of predispose to hypertension in Kazakans. Therefore we investigate the relationship between the Gly460Trp polymorphism of α -adducin gene and essential hypertension in Kazakans of Xinjiang. Methods We recruited 367 Kazakans subjects (235 hypertensives and 132 normotensives ) from Aletai, Tacheng and Xinyuan pasturing areas in this population-based case-control study. Samples of peripheral blood were collected from all recruited subjects. The DNA was extracted by means of a standard phenol-chloroform method. Mutagenically separated polymerase chain reaction (MS-PCR) methods were used to detect the Gly460Trp polymorphism of a -adducin gene. Results The Gly/Gly, Gly/Trp, Trp/Trp genotype frequencies of α -adducin gene in Kazakans of Xinjiang were 23. 83%, 50.64%, 25. 53% respectively; 460Gly and 460Trp allele frequencies were 49. 15%, 50.85% respectively in essential hypertension group. While the corresponding frequencies were 26. 52%, 46.96%, 26.52%; 50%, 50% respectively in normotensive group. There was no significant difference in 460 Trp/Trp genotype and 460Trp allele frequencies of a -adducin gene between hypertensive and normotensive group (P>0. 05) . Systolic blood pressure, diastolic blood pressure and body mass index had also no significant difference in the subgroups classified according to genotypes (P>0. 05) . In addition, there was no significant difference in 460 Trp/Trpgenotype frequencies of a -adducin gene between hypertension and normotensive group in the subgroups classified according to body mass index (P>0. 05) . Conclusion The 460Trp variant of α-adducin gene is not association with essential hypertension in Kazakans, which implicates it may not play a important role in the pathogenesis of hypertension in Kazakans of Xinjiang. |
