Objective: The gastric carcinoma is one of the familiar malignant tumors in clinic, having all malignant tumors death of 23.03%.The tumor cells invasion-metastasis and recurrence after surgery are the major reason for the poor prognosis of the patients with gastric carcinoma. To evaluate the effects of vascular endothelial growth factor expression on invasion and metastasis of gastric carcinoma,their regional lymph and their relationship with clinic pathologic features. To evaluate its role in the estimation of the tumor progression, metastasis as well as prognosis. Methods: Select 62 gastric carcinoma patients who had operations for radical cure in surgical department three of the HeBei Medical University Forth Hospital Department, and they were all proved by pathology. Collect 62 cases of gastric carcinoma tissues ,62 cases of regional lymph,30 cases of normal gastric mucosa in operating room. The samples of gastric carcinoma, regional lymph node and normal gastric epithelia were fixed by 10% neutral formalin, dehydrated by alcohol and embedded by paraffin. Routine diction of 4μm thickness for 3 sections and 1 section was made H.E staining. Pathology diagnosis were made by two experienced pathologist. The sections were subjected to antigen retrieval in boiling sodium citrate buffer (0.01M,PH6.0)for 10min under microwave. SP immunohistochemical staining was used for VEGF ,bFGF detection in carcinomas ,lymph node metastasis tissues and normal gastric carcinoma. The relationship between the expression of VEGF ,bFGF and the clinical biological characteristics, including age,sex,tumor location ,size,Borrmann type ,differentiation,invasion depth, lymph node metastasis ,and TNM stage,5-year survival. The sections were subjected to sequential 3,3-diamino-benzidine(DAB) for immune complex visualization and then counter stained with haematoxylin. All statistical analyses were carried out with SPSS 10.0, p<0.05 was considered significant. Results:1 Expressions of VEGF,bFGF in tissues of gastric carcinoma were significantly higher than in normal gastric mucosa .The results indicated that expression of VEGF,bFGF in tissues of gastric carcinoma increased abnormally. There is statistical significance (p<0.05). 2 There were no significant correlation between VEGF positive expression in gastric carcinoma tissues and the sex,age,tumor location ,size,differentiation ,lymph node metastasis ,Borrmann type of the patient respectively. The positive rate in the stage Ⅲ-Ⅳgroup were significantly higher than in the Ⅰ-Ⅱg roup(p<0.01). The positive rate in the stage T3-T4group were significantly higher than in the T1-T2 group(p<0.01). The positive rate in the stage 5-year survival <5 group were significantly higher than in the 5-year survival ≥5 group(p<0.01). There is statistical significance (p<0.01). 3 There were no significant correlation between bFGF positive expression in gastric carcinoma tissues and the sex,age,tumor location ,size,pathohistological type ,Borrmann type,lymph node metastasis ,invasion depth of the patient respectively (p>0.05).The positive rate in the stage Ⅲ-Ⅳgroup were significantly higher than in the Ⅰ-Ⅱgroup There is statistical significance (p<0.05). The positive rate in the stage 5-year survival <5 group were significantly higher than in the 5-year survival ≥5 group(p<0.05). There is statistical significance (p<0.05). 4 In gastric regional lymph nodes, the expressions of VEGF and bFGF were significantly higher with lymph node present group than those without lymph code metastasis one . There is statistical significance (p<0.01). 5 In gastric regional lymph nodes, the lymph node metastasis rate in positive expression group of VEGF and bFGF were significantly higher than that in negative one. There is statistical significance (p<0.01). 6 In gastric regional lymph nodes,5-year survival rate in positive expression group of VEGF were significantly lower than that in negative one(p<0.01).5-year survival rate in positive expression group of bFGF were significantly lower than thatin negative one(p<0.05). conclusions: 1 Expressions of VEGF and bFGF in gastric carcinoma tissues were abnormal .To detect VEGF and bFGF would be useful molecular marker for gastric diagnosis. 2 Expressions of VEGF in gastric carcinoma tissues had no correlation with the patients sex,age,tumor location ,size,pathohistological type,Borrmann type,lymph node metastasis. It expression was correlated with TNM stage,invasion depth and 5-year survival rate significantly. All that indicate there is correlation between the three indices and the attack,metastasis or development of gastric carcinoma, to test the concentrations of VEGF in gastric carcinoma tissues will help to judge metastasis,recrudescence and monitor prognosis. 3 Expressions of bFGF in gastric carcinoma tissues had no correlation with the patients sex,age,tumor location ,size,pathohistological type,Borrmann type,stage,TNM stage lymph node metastasis. It expression was correlated with invasion depth and 5-year survival rate significantly. To test the concentrations of bFGF in gastric carcinoma tissues will help to judge metastasis,recrudescence and monitor prognosis. 4 In gastric carcinoma and in the regional lymph nodes, the high expression of VEGF and bFGF could be important factors affecting prognosis of gastric carcinoma patients, so the detection of their expressions may be useful to prognosis of gastric carcinoma.
