[Background and Objective]:Growth tumor and the formation of metastases are dependent on the formation of new blood vessels. Angiogenesis is an absolute requirement for the neoplastic growth of solid tumors after they reach a critical size of 1-2mm~3. Tumor cells have been shown the secrete a variety of angiogenic factors and thereby induce the local formation of new blood capillaries. Among the angiogenic factors, the vascular endothelial growth factor(VEGF) play a pivotal role. Most solid tumors express high levels of VEGF, including breast, gastric and colonic carcinormas.Carcinoma of Stomach the disease that severely harmed to peple's health,its incidence of disease has primacy in digestive system malignancy .In our country , the death rate was 25.16/10~5,occupied 23.02% of malignancy's death rate and as the primacy in all kinds of carcinoma. But the checkout rate of early Carcinoma of Stomach less 10% in clinic , most operation patients were advanced carcinoma,its mainly metastasis approach had by the way of lymphatic , therefore, the mostly factor that affected patient prognosis was lymph node metastasis, the molecule and biology mechanism of lymphatic metastasis in carcinoma of stomach had not clarified. VEGF-C and VEGF-D as lymphatic building gene,could combined with VEGFR-3 that expressed in lymphatic endothelial cells, induced to build , hyperplasia and dilatation of lymphatic, accordingly formed a way that accelerated tumor cells metastasis to lymph node. Checked the expression of VEGF-C and VEGF-D in tissue of carcinoma of stomach had been used as to realize the relationship between VEGF -C ,D and invasion and metastasis of carcinoma of stomach . In this study ,we would survey the expression of VEGF-C and VEGF-D in serum and tissue with advanced gastric carcinoma to clarify the Correlation between it's and lymph node metastasis .[Methods] :1.Enzyme linked immunosorbent assay (ELISA) were used to detect serum VEGF-C and VEGF-D concentration in 88 cases of human AGC , 10 EGC and 10 health controls.2.Using immunohistochemical staining (EnVisionTM method) To determine the expression of VEGF-C and VEGF-D in these kinds of tissues.[Result]:1.The serum level of VEGF-C and VEGF-D in AGC was higher obviously than controls group(P<0. 05 ) but have no significance with EGC(P>0. 05).2.The high serum level of VEGF-C in AGC was correlated with lymph node metastasis(P<0.05).The serum level of VEGF-D in AGC was not correlated with lymph node metastasis(P>0. 05).3.The positive expression of VEGF-C and VEGF-D was 52.5% and 62.5% respectively in AGC ,The postive expression in AGC was much higher than in EGC and health controls.4.The positive expression of VEGF-C in lymph node metastasis group higher remarkably than had no lymph node metastasis group and had statistics significance. Compared with had no lymph node metastasis group ,the positive expression of VEGF-D had no statistics significance in AGC (P>0.05).[Conclusion] :The serum level of VEGF-C had potential usefulness as biologic markers of Advanced gastric carcinoma; The expression of VEGF-C,D was highly in the tissue;The tissue expression of VEGF-C is closely related to lymph node metastasis of AGC; But VEGF-D have no significance correlation.
