Objective:Using MTT and flow cytometry we investigated the effect of reversing multidrug resistance (MDR) in leukemia cells by Angelicasinensis (Oliv) Diels compound Injection, Brucine, Strychnine , and explored the underlying mechanisms of these effects.Method:Cell lines K562/A02 and K562/S were treated with Angelicasinensis (Oliv) Diels compound Injection, Brucine, Strychnine respectively. Cytotoxic effect of these agentswas analyzed by MTT and Trypan blue. After the cells had been treated with these agents individually at the dose level of IC10, or in combination with ADM, expression of P-gp and DNR concentration in the cells were analyzed by flow cytometry.Results:1, After being treated with these Chinese traditional medicine, the multidrugresistance of K562/A02 cells were partially reversed. IC50 values of K562/A02 cell line, which was affected by ADM, were reduced. Thus the sensitivity of target cells to ADM was upregulated.2, When each of the agents were administered individually, the P-gp expression inK562/A02 cells could be reduced. When combined with ADM, these agents markedly reduced the expression of P-gp.3, The tested agents improved the accumulation of medicine in K562/A02 cells.Conclusions:1 , Angelicasinensis (Oliv) Diels compound Injection, Brucine, Strychnine could partially reverse MDR in leukemia cells.2, These agents reduce MDR by improving drug concentration in leukemia cells and affecting P-gp pump.
