ObjectiveGlioma is one of the most common tumor of nervous system. Its surgical resection rate is low, recurrence rate is high and survival rate is low. The kind of malignant biological behavior is determined by the invasive growth manner.There are lots of factors correlated with human brain gliomas invasion, a-mong which, neural cell adhesion molecule (NCAM) ,matrix metelloproteinases (MMPs) and tissue inhibtors of metalloproteinases (TIMPs) are most colsely related to gliomas invasion.The study examined the expression of NCAM -1, MMP - 9 and TIMP - 1 in normal brain tissue and gliomas of various grades by immunohistochemical te-chiques, analysed their pathological index ( PI) , which aimed to probe into the role of NCAM, MMPs and TIMPs in human brain gliomas invasion and their relationships.Materials and methodsThe study selected 40 cases of gliomas that received surgical resection or biopsy. There were 22 male and 18 female, with a median age of 46 years (ranging 12-76). The subjects were classifed into four grades according to the pathophysiological criteria of WHO(1999) ,in which,8 cases were grade Ⅰ ,14 cases were grade Ⅱ, 12 cases were grade Ⅲ, 6 cases were grade Ⅳ; Ⅰ - Ⅱ grade were named low - grade gliomas ( LGG) group and Ⅲ - Ⅳ grade were named high -grade gliomas (HGG) group. 6 cases were normal brain tissues.Specimens were fixed with formalin and embedded with paraffin, made4μm thick section. Immunohistochemistry staining (S - P method) was used to detect the espresssion of NCAM -1 ,MMP -9 and TIMP -1 in two groups.Results determination criteria: In microscope cells presenting brown particles in nucleus or plasma were regarded as positive cell. The proportions of positive cell were beyond 5% that were regarded as positive, otherwise, were negative. Pathological index (PI) was used to count positive cell, PI value was treated with semi - quantitative method.Statistical analysis: SPSS statistical software was used for statistical analysis. The comparison between group positive rate was examined by X2 test. The comparison between group PI value examined by t - test. The relationship of two factors was examined by linear regression. The level of significance was set to ( P < 0.05).Results1. The expression of NCAM - 1: The expression of NCAM - 1 in normal brain tissue was negative. The expression of NCAM -1 in gliomas was located in tumor cell plasma and nucleus. The expression positive rate of LGG group was 77.27% (17/22 ) , PI value 32. 77 ± 10. 77, the expression positive rate of in HGG group was 33.33% (6/18) ,PI value 19.72 ±7. 14,significant difference between two groups ( P < 0.01)2. The expression of MMP -9: The expression of MMP -9 in normal brain tissue was negative. The expression of MMP -9 in gliomas was located in tumor cell plasma, messenchymal cell and extracelluar matrix(ECM) ,most of expression in tumor cell of invasive border and vascular basal membrane epithelial cell. The expression positive rate of LGG group was 59.09% (13/22) ,PI value 25.54 ± 10.12,the expression positive rate of in HGG group was 100% (18/ 18) ,PI value 40.48 ±7.26, significant difference between two groups (P<0. 01).3. The expression of TIMP -1: The expression of TIMP -1 in normal brain tissue is negative. The expression of TIMP -1 in gliomas is located in tumor cell and ECM, most in tumor cell.
