ObjectiveTo investigate the influence of acupuncture on the relevant indexes in rats with diet-induced obesity, including serum leptin, body weight, body length, Lee' s index, the quantity of fat in pericardium, perirenal area, and epididymis.MethodsIn the research, 80 weaning male SD rats were randomly assigned into normal diet group, high-fat diet with acupuncture group and high-fat diet group. The models were established after 3 months. In the experiment, the rats were divided into 6 groups: normal diet group (6 rats), high-fat with acupuncture group (6rats), obesity and normal diet group (8rats), obesity and high-fat-diet group (8 rats), obesity with high-fat-diet and acupuncture group (8rats). During the experiment, the rats in different groups were fed with the relevant feeds, got the feeds and water freely. The feeds and water were changed every day. The rats were put in the fixer to adapt for 15 minutes daily for 14 days. Rats in the acupuncture group started to receive acupuncture on the 15th day at "Housanli" and Neiting (ST44). No. 36 cosmetic needle were used to puncture the two points 4mm and 2 mm deep individually. Even reinforcing and reducing methods were applied at Housanli for 1 minute. Reducing method by twirling the needle was applied at Neiting for 0.5 minute. The electroacupuncture machine was used for the two points with continuous wave with 10 Hz and 1. 5V. The treatment had been conducted for 10 minutes daily for 14 days. The points at each side of the rats were used alternatively. The treatment in the six groups were arranged at 18:00-20:00 daily for 14 days. During the experiment,the amount of food and water intake, urination, bowl motion, the body weight, and body length were measured. The Lee' s index was then calculated. The rats were in fasting overnight. When the experiment finished, the rats were decapitated in the morning of the next day. The fat of the pericardium, perirenal area and epididymides were separated to measure the weight. During the execution of the rats, 5ml of blood was collected rapidly and put into anticoagulation tube with 0.1% heparin, centrifugated to take the serum, and put in - 20°C low-temperature refrigerator for reservation. The serum was measured by radioimmunoassay. The other were measured by routine methods.Results(1)Comparing with the normal diet group, the body weight and Lee' s index in the high-fat diet and acupuncture group did not increased significantly (P>0. 05). The difference had no statistical significance. Comparing with the relevant non-acupuncture groups, i.e. the obesity normal diet group and the obesity high-fat group, the body weight and the Lee' s index of the obesity normal diet acupuncture group and the obesity high fat acupuncture group decreased significantly (P<0.001, 0.001, 0.01, 0.001). The difference was significant. The Lee's index and body weight in the obesity high-fat acupuncture group were higher than those in the obesity normal acupuncture group, but P>0.05 which had no statistical difference. (2) The quantity of fat in the pericardium, perirenal area and epididymides in the high-fat acupuncture group had no difference comparing with the normal diet group (P>0. 05). The difference has no statistical difference. Comparing with the non-acupuncture groups, i. e. the obesity normal diet group and the obesity high fat groups. The quantity of fat in the pericardium, perirenal area in the obesity normal diet acupuncture group and the obesity high-fat acupuncture group decreased significantly (P<0. 001, 0.05, 0.05, 0.01). The difference was significant. The quantity of fat at the epididymides decrease significantly in the obesity normal diet acupuncture group comparing with the obesity normal diet group (P<0. 05), but that in the obesity high-fat acupuncture group had nodifference with obesity high fat group (P>0. 05). This may be due to the quantity of samples. The quantity of fat in the peridium, perirenal areas, and the epididymides in the obesity high-fat acupuncture group was higher than that in the obesity normal diet acupuncture group. P<0.05 in the pericardium with significant difference. P>0. 05 in the perirenal area and epididymides without significant difference. (3) Comparing with the normal diet group, the quantity of the serum total cholesterol and triglyceride in the high-fat acupuncture group did not increase. Comparing with the two, P>0.05 without statistic difference. Comparing with the relevant non-acupuncture groups, the triglyceride in the obesity normal diet acupuncture group and obesity high-fat acupuncture group reduce significantly (P<0.05, 0.01). The quantity of the serum total cholesterol comparison had no statistic difference (P>0. 05), which may be due to the quantity of samples. (4) The quantity of the serum leptin has no difference comparing with that in the normal diet group. Comparing with the relevant non-acupuncture group, the serum leptin in acupuncture groups did not reduce significantly, p>0. 05, which may be due to the quantity of samples. The serum lepin in the obesity high-fat acupuncture group was higher than that in the obesity normal diet acupuncture group with the P value higher than 0. 05 which has no statistical significance.ConclusionThis research showed that acupuncture relieve the high serum leptin state of the body, reduce the leptin resistance, and promote its biological effect by affecting the food intake, increasing the body energy metabolism to relieve obesity. High-fat diet has important promoting effect on simple obesity. Acupuncture can improve the lipid metabolism, reduce blood TC and TG, reduce the blood viscosity and improve the arteriosclerosis index (AI) and prevent heart and cerebrovascular diseases effectively. Acupuncture with dietary control (controlling the intake of high-fat food) has good effect to reduce obesity index, quantity of fat in pericardium, perirenal area, epididymides, and the serum total cholesterol, triglyceride quantity.
