Objective: To investigate the effects of transforming growth factor-Pi (TGF-β1) in the proliferation and collagen synthesis of human orbital fibroblasts ( OFs) cultured in vitro and to explore the contribution of TGF-β1 in proliferative vitreoretinopathy (PVR) formation . Methods: With the cultured human orbital fibroblasts in vitro from the man whose eye globe has been removed because of ocular trauma and want to implant the artificial eye. The orbital scarring tissue has been planted in the culture flask to obtain the primary culturing orbital fibroblasts , Different concentration of TGF-β1(0.1, 1.0, 5.0, 10.0, 100.0μg/L) were incubated with subculturing human orbital fibroblasts. The cell growth curves were drawn to observe the cell proliferation, trypan blue and MTT assay was used to determine the proliferation activity of OFs, The cell proliferation was determined by ~3H-TdR absorption, The change of cell cycle were observed by Flow cytometry (FCM), while the collagen synthesis was evaluated by ~3H-Proline incorporation.Result: TGF-β1 of (0.1,1.0,5.0,10.0μg/L) was significantly increasing the proliferation and collagen synthesis of human orbital fibroblasts.
