Objective To study the effects and pharmacological mechanism of Tuo Min Zu Fang (TMZF) in treating experimental hypersensitivity type I in vivo and in vitro. To supply some pharmacological basis for the application of TMZF in treating allergic diseases clinically.Methods (1) In vivo experiment: 1)anti-serum of rats administrated with ovalbumin was prepared first. 2)in passive cutaneous anaphylaxis (PCA), the experimental model of hypersensitivity type I were induced by intradermal injection of rat anti-ovalbumin antiserum on normal rats, the experimental models were divided into five groups and feeded with the corresponding drugs: the normal group(NS), the control group(NS), the treatment group of TMZF in two concentrations: 1.4g/ml, 2.8g/ml and the treatment group of chlorphemramine ( 4mg/kg) , then they were attacked by ovalbumin and Evans' blue. The effusion of Evans' blue from the skin being passive allergized were determined spectrophotometrically at 610nm, and the inhibition ratio to PCA were used to show the effect of TMZF to hypersensitivity type I. 3)We observed the morphologic changes of mast cells in rat calvarial periosteum, which were also allergized by anti-ovalbumin antiserum to study the effects of TMZF in stabilizing the membranes of mast cell in vivo. (2) In vitro experiments: rat peritoneal mast cells(RPMC) were isolated through centrifugation on density gradients of Percoll. The cell models were pre-incubated with anti-ovalbumin antiserum in vitro to be elicited. To examine TMZF' effects of stabilizing the membranes of mast cells inhibiting degranulation of mast cells and decreasing the setting off of mediators of inflammation from mast cells, we did the following experiments: 1)oluidine blue was used to stain the cell models to observe the morphologic changes after treated with TMZF. 2)The influence of TMZF on histamine release from RPMC elicited by anti-ovalbumin antiserum were examined by fluorescence...
