Objective: To study the effects of indocyanine green (ICG) on proliferation by rabbit lens epithelial cells (RLECs) in vitro,and to investigate the safety and reliability of the technology of staining of the lens casular with ICG during cataract surgery. Methods: RLECs were cultured in vitro. 1.The effects of ICG at different concentrations on proliferation of RLECs in vitro at 24h, 48h, 72h was measured with MTT staining colorimetrey and cell count technique; 2. The morphological of RLECs changes were observed with light microscope.Results: 1.The result of MTT showed that ICG at the concentration of 0.5% and 1% can significantly promote the RLECs proliferation at the time of 24h (P<0.05). At 48h and 72h the effects of the proliferation on RLECs of ICG at all of the experimental concentrations were significant (P<0.05). At the same time the effects of proliferation were depended on concentrations (P<0.05). 2. The result of cell count technique at 24h was consistent to that of MTT. With the prolongation of the time the adheration of the RLECs was significantly increased, the RLEcs can't be separated, so the cell count can't be implemented at 48h and 72h. 3. Observation of the morphological changes with light microscope:normally cultured RLECs have the characteristics of the epithelial cells, the RLECs used by ICG showed that the plasma was relatively little, the membrane of the nucleole was clear, nucleole was obvious, the ratio of nucleole and plasma was bigger. Conclusions: ICG can promote the RLECs proliferation in vitro,and can contribute to the posterior capsular opacification by this way. In order to decrease the occurence of PC0,we should use ICG carefully during the cataract surgery.
