Objective To investigate the expression of intercellular adhesion molecule 1(ICAM-1) and the changes of ICAM-1 in cerebra after focal cerebral ischemia and reperfusion, to study the effects of inosine on the expression of ICAM-1 in cerebra and to explore the neuroprofectivity of inosine.Methods some adult female Sprague Dawley(SD) rats were divided randomly into sham operation group(SG),ischemia-reperfusion group(IG) consisted of 7 subgroups with 3 rats and therapy group(TG) consisted of 7 subgroups with 3 rats, and they were made to Middle Cerebral Artery Occlusion(MCAO) model at 2h,6h,12h,24h,2d,3d,7d and 14d respectively after ischemic 90min. the expression of ICAM-1 and its mRNA were measured by methods of immunohistochemistry(IHC) and in site hybridize(ISH).Results At the IG The level of ICAM-1 in cerebral contex and striaturn varies with the hours of ischemia reperfusion, and the eyeable expression of ICAM-1 and its mRNA occur at 2h possibly, a marked increase at 6h, the peak at 24h, the heaps remained from 12h to 48h and then decrease gradually, whereas they were observed hardly in the SG; At the TG, in despite of the level of ICAM-1 and its mRNA also varies with the hours of ischemia-reperfussion, the number of cerebral positive cells are fewer than corresponded hour in the IG and higher notable than the SG There is a significant difference between the IG and the TGConclusion appropriate concentrate of inosine might be beneficial to cerebral tissue after ischemia reperfusion.
