Objective: In this study , by observing the count of white blood cells (WBC) orCD34+ cells in peripheral blood and side effects by GranulocyteColony-Stimulating Factor (G-CSF) in patients with acute myocardialinfarction (AMI), we evaluated the mobilization effects, the safety and thefeasibility of autologous peripheral blood stem cell by G-CSF in patientswith AMI. Method: Patient Selection: There are total 44 patients with AMI in our hospital attending thisstudy .The institution ethics review board of Liaoning province people'hospital approved the study protocol. We obtained informed writtenconsent from patients after explaining the procedure and risk. Patientsbelow 75 years of age were eligible for inclusion into this study if they hada first acute ST-elevation during 1 month that was diagnosed as AMI andwas treated by coronary stenting where the culprit lesion of theinfarct-related artery. The following exclusion criterias were required forpatient enrollment: (1) difficulty in obtaining vascular access forpercutaneous procedures or ineligibility for percutaneous revascularization;(2) a history of tumor or disease affecting survival in short time; (3) age>75;(4)anemia(Hemoglobin﹤100g/L);(5) active infection; (6) bone tissuewith abnormal radiological aspect;(7) the presence of coagulationdysfunction;(8) the presence of cardiogenic shock (defined as systolicblood pressure﹤80mmHg);(9) any condition that, in the judgment of theinvestigator ,would place the patient at undue risk. All patients were placedon maximally tolerated medical therapy at the time of enrollment. Method and Observed target: 44 patients with AMI receiving standard drug and coronary arteryintervention therapy(being treated by coronary stenting where the culpritlesion of infarct-related artery), were randomly allocated to three groups.One (A group) was received G-CSF 300μg /day, another (B group) wasreceived G-CSF 600μg /day, and the third one (C group) was not receivedG-CSF. The patients were received G-CSF, by hypodermic injection, andthe duration of applying G-CSF was 5 days. In the process of themobilization of the peripheral blood stem cell, prior to applying G-CSF andon the 3rd,4th,5th,6th,7th after applying G-CSF ,the white blood cells(WBC) in the peripheral blood should be observed, and on the 3rd,4th,5th,6th,7th after applying G-CSF ,CD34+ cells count in the peripheralblood should also be observed ;and the following side effects also shouldbe paid attention to, such as: bone pain, tetter, fever, gastrointestinaleffects( nausea,vomit,coprostatis ),deteriorated of angina or heart failureand some rare effects(spontaneous spleen rupture , severe purulentinfection,hypercoagulable state,autologous immune diseases). Due to thethird group ,which was not received G-CSF ,on the 3rd,5th,7th,8th afterAMI, CD34+ cell count in the peripheral blood should be paid attention to. Statistics: Continuous numerous variables were expressed as mean value ± SD.SPSS 10.0 statistics software were adopted. Differences in variables of Agroup, B group and C group were assessed by one-way ANOVA;differences in variables of A group and B group were assessed byIndependent Sample T Test; count variables were assessed by Chi-Square.A value of P<0.05 was considered statistically significant. Results: 1.Due to A group, prior to applying G-CSF and the 3rd,4th,5th,6th,7th after applying G-CSF, the counts of WBC were (7.74±1.90)×109/L,(28.76±7.00)×109/L,(35.24±9.38)×109/L,(31.47±10.03)×109/L,(46.32±63.77)×109/L,(15.75±10.04)×109/L;on the 3rd,4th,5th,6th,7th after applying G-CSF, the counts of CD34+ cells were(55.47±26.51)×106,(84.43±102.07)×106,(98.99±82.08)×106,(90.46±70.90)×106,(36.08±29.95)×106. 2.Due to B group, the counts of WBC were(7.83±2.83)×109/L,(42.99±21.12)×109/L,(43.08±21.01)×109/L,(46.46±21.89)×109/L,(91.52±125.98)×109/L,(23.51±9.56)×109/L;the counts of CD34+ cellswere (96.60±40.07)×106,(157.02±131.84)×106,(330.21±203.36)×106,(264.60±165.32)×106,(86.25±43.9...
