Objective: to investigate the frequency of antibody response of SMP-30 and its specificity in patients serum of hepatocellular carcinoma, other tumors, nontumorous liver disease, other nontumorous diseases and health adult control.Methods: with indirect enzyme-linked immunosorbent assay(ELISA) and western blot the frequency of SMP-30 antibody in serum from 145 patients with HCC, 366 patients with other tumors, 79 patients with nontumorous liver disease, 20 patients with nontumorous diseases and 138 health adult control were tested with purified SMP-30 fusion protein recombined and expressed by the technique of genetic engineering. The results were analyzed by SPSS statistic analysis software.Results: the principal frequency of antibody against SMP-30 in serum from HCC patients is 32.4%(47/145), there is only 0.72%(1/138)in health adult control group, statistic analysis show great significance (p<0.001) between the two groups. The results show that 13.3%(13/98)of nasopharyngeal carcinoma(NPC), 7.7% (2/26) of esophageal carcinoma, 2.5% (1/40) of intestines cancer, 7.3% (3/41) of cancer of the cervix patients in other tumors group and 6.6% (4/60) of type B hepatitis, 5.3% (1/19) of hepatic cirrhosis in nontumorous liver disease group have antibodies against SMP-30,and that shows great significance compared with HCC group, too. In addition , the positive rate of SMP-30 could reach 43.6%(17/39) among the HCC patients with Alpha Fetal Protein(AJFP) negative in the serum, which hints that it is a complementarity for the diagnosis of HCC. The sensitivity, specificity, accuracy, positive predictive value, negative predictive value, positive likelihood ratio and negative likelihood ratio of SMP-30 in the diagnosis of HCC are 32.4%, 95.9%, 83.7%, 65.3%, 85.6%, 7.9and 0.71, which indicates to possess a magnitude value in HCC auxiliary diagnosis with serology engineering.Conclusion: with certain specificity in HCC and complementarity for the diagnosis of HCC in patients with AFP negative, antibody against SMP-30 detection might be a new method in HCC auxiliary diagnosis with serology engineering.
