Corydalis decumbentis (Thunb.)Pers. is a herb of promoting blood circulation, removing collaterals in the channels and promoting the circulation of Qi to relieve pain, used for the treatment of paralytic stroke, traumatism, rheumatic arthritis and sciatica. Comprehensively various methods of isolation and structures identification were employed,studies on chemical constituents of Corydalis decumbentis were carried out more deeply and systematically.From rhizoma of Corydalis decumbentis, thirteen constituents were isolated by combination of silica gel column chromatography , gel electrophoresis column chromatography, preparative TLC.On the basis of their chemical and physical properties as well as the various spectra data of 1D-NMR, 2D-NMR, ESI-MS, IR et al, combined of abundance of literatures, the structures of twelve constituents were determined, they are P-sitosterol(â… ), (+)-tetrahydropalmatine(â…¡), kikemanine(â…¢), bicuculline(â…£), protopine (â…¤), a-allocryptopine(â…¥), daucosterol(â…¦), muramine(â…§), bulbocapnine(â…¨), bitetrahydro-isoquinoline(â…©), two isoquinoliums(â…ª, â…«).Among them, â…¨ was isolated from this plant for the first time, while â…©, â…ª and â…« were three new compounds from natural products, which have a close relation with isoquinoline, and they may be evolved from isoquinoline on biogenic relationship.In the primary quality standard study of Corydalis decumbentis, selecting protopine as study objects, using C18 column, acetonitrile-1% ethyl acetate( 25:75) (PH.6.0 adujusted bytriethylamine) as mobile phase, detection wavelength at 290nm, determination method of protopine in Corydalis decumbentis was developed by HPLC. Using Agligent UV-vis spectrophotometer, detection wavelength at 525nm, determination method of alkaloid extract in Corydalis decumbentis was developed by colorimetry. These two methods are simple and accurate and have good precision and ruggedness, and can be used to control the quality of Corydalis decumbentis and its preparation.Effect of alkaloid extract and bicuculline of Corydalis decumbentis on arrhythmia of Wistar mice was determined by mice antiarrhythimia model. Lidocaine was being employed as the positive drug, results show the time of ventricular extrasystole(VE) and ventricular tachyardia(VT) in the lower-dose of alkaloid group was markedly prolonged, the concentration is 7.5mg/kg; but in the higher-dose group it was not remarkable, just trended to be prolonged. Bicuculline group could remarkablely prolong the time of VE.It has significant difference between bicuculline group and lidocaine group(P<0.01).The time of VT is trended to be prolonged,not remarkable, the concentration of bicuculline is 30mg/kg.Studies on chemical constistuents of Corydalis decumbentis were carried out more deeply and systematically. Twelve constistuents were isolated and determined, among which â…¨ was isolated from this plant and â…©, â…ª, â…« were three new compounds. In the primary quality standard study of Corydalis decumbentis, determination method of protopine in Corydalis decumbentis by HPLC and determination method of alkaloid extract in Corydalis decumbentis by colorimetry were established. Also the antiarrhythmia activity of alkaloid extract in Corydalis decumbentis was evaluated, and the antiarrhythmia activity of bicuculline was found for the first time. This thesis will provides powerful basis for deep studies of Corydalis decumbentis and quality standard of Chinese traditional medicine exploitation.
