Objective:The ultra-fine powder technology has been widely useded in traditional Chinese medicine industry.The active ingredients of Chinese herbal medicine are usually distributed in between the cytoplasm of the cell and cells,and most of them are in cells.Superfine powder technology is introduced to Chinese medicine field.However,the application of superfine powder in Chinese pharmaceutical industry is still at the exploratory stage.We should perfect the powder quality standardization system,and strengthen the systematic study of the powder preparation process technology,quality standards and the major pharmacodynamics.In this thesis,as the research object of Corydalis Decumbentis,Rhizoma Study on superfine grinding of Corydalis Decumbentis Rhizoma,the fingerprint of the superfine of Corydalis Decumbentis Rhizoma,Study correlation on its analgesic effect of the alkaloids and the blood contained in the amount of alkaloids.which provide experimental data for formulating and perfecting the quality standards of Chinese medicine ultrafine of Corydalis Decumbentis Rhizoma.Methods:(1)Study on superfine powder of Corydalis Decumbentis Rhizoma grinding time.By research at the different time-crushed powder dissolution of extraction,and study on dissolution of protopine,palmatine.(2)Study on characterization and physical properties of superfine powder of Corydalis Decumbentis Rhizoma:To examine particle size detection,Microstructure,powder bulk densitty and other aspects of powder of Corydalis Decumbentis Rhizoma.Examine the mobility of the different particle size powders and hygroscopic change by through the angle of repose determination as well as moisture absorption rate,moisture percentage,the relative critical humidity.(3)Study on fingerprint of ultra-fine powder of Corydalis Decumbentis Rhizoma.As protopine,Tetrahydropalmatine,palmatine to the targets.To establish the fingerpint of the ultra-fine powder.(4)Study correlation on Ultra-fine grinding before and after Analgesic effect and plasma concentration of Corydalis Decumbentis Rhizoma.Study correlation by acetic acid induced writhing response analgesic effect in mice and the blood contained in the amount of alkaloids.The piece of Corydalis Decumbentis Rhizoma after the ultra-fine powder improve bioavailability,clinacal science and rational drag use,provide a certain amount of experimental evidence.Result:(1)Study on superfine powder of Corydalis Decumbentis Rhizoma grinding time.After 18 minutes of grinding time,The indicators without increase of grinding time and the significant changes.(2)Study on characterization and physical properties of ultra-fine powder,After ultrafine grinding,The microscopic identification organelles appear is broken.And the lack of microscopic identification of common characteristics.Powder's texture tends to be uniform,the mobility decrease,the moisture absorption increases.(3)Establish 10 batches of different origin HPLC fingerprint of ultra-fine powder,Established 11 common peaks.Similarity of 10 batches of samples above 0.90 specific.HPLC fingerprints of control the ultra-fine powder are extracted with methanol and the inherent quality of ultra-fine powder.(4)By ultrafine grinding of Corydalis Decumbentis Rhizoma,Stronger inhibitory effect on acetic acid induced writhing response than the ordinary pieces,The main Alkaloid,protopine Tetrahydropalmatine,palmatine content in the blood is higher than ordinary pieces.The analgesic effect and the plasma concentration of brewing group and returning gronp is similar.In order to improve the bioavailability of Rhizoma Corydalis Decumbentis,reducing the dose of the clinical applications to facilitate the patient taking,Promote the transformation and use of traditional herbs,with reference to the role.Conclusion:After ultrafine grinding,The particle size distribution,microscopic characteristics,moisture absorption,and other physical characteristics are varying degrees of change,Pieces of Corydalis Decumbentis Rhizoma ultrafine grinding analgesic effect and the blood before and after shows,The same dose of analgesic effect of ultra-fine powder of analgesic effect is significantly stronger than ordinary pirces.Supermicro crushed its efficacy play a positive role to prove the superiority of traditional medicine superfine powder.
