Background and Objective: Transitional cell carcinoma of the bladder is themost common neoplasm of urological system.It takes about 20% of all the malignant tumour and the incidence of it is situated on the first in urological system tumour. The incidence is70% in people over 50,and 85% of the patients are males.The key cases caused the death of transitional cell carcinoma of the bladder are its invasion and metastasis.So the study about pathogenesis,development,invasion and metastasis of transitional cell carcinoma of the bladder has become a focus.Recently,great attention has been paid on the relation between angiogenesis and the pathogenesis,development of the tumour.Not only the new blood vessels bring nutrition to the tumours,but theyalso give the tumour cells chances to be transferred. A great deal of studies show that there is a high relationship between angiogenesis and tumoue'smetastasis.The microressel density(MVD)of the tumoue tissues not only reflect the growth of the tumour blood vessels quantitatively ,but can also predict the progress,metastasis and recurring of the tumour.CD34,CD105 are ofen used to be the markers of MVD,the CD 105 is one of the glycoprotein of the cell membrane belonging to β super family of transforming growth factor.Recently,some studies show that in the tumour tissues ,the expression of CD 105 is very high in the endothelial cells which proliferate actively.but there is no expression in normal tissues.For that the expeession of CD105 in the tumour tissues can reflect the angiogenesis of the tumour and the actively of the endothelial cells,we can take it one of the indicators to judge metatasis and predict the prognosis.There are some studiesshow that CD105 not only takes part in the pathogenesis and development of breast cancer,esophageal carcinomar,the more important is that it can also play roles in the invasion and metastasis.CD34 is a phosphate protein belonging to transmembrane protein-I and is mainly expressed on henatopoietic stem .Reserchs have approved that the CD34 in the normal marrow and peripheral blood was closed correlated with the hematopoiesis,great deal of reserchs have demonstrated that CD34 can be a marker of endothelial cells of the big blood vessels ,but the stain is very faint in endothelial cells of many tumour tissues,furthermore ,the expression of CD34 or factor VIII is usually used to be the marker of MVD,there is little research which sign MVD with CD105,There is no report on taking CD34 and CD 105 simytaneously as markers of MVD to research their correlation in transitional cell carcinoma of the bladder tissues.In this study,by adopting immunohistochemisty sp method,detecting the expression of CD34 and CD105 in the transitional cell carcinoma of the bladder and analyzing their correlection,then discussing their roles in the pathogenesis, development, invasion and metastasis in transitional cell carcinoma of the bladder tissues and discussing their correlation in the invasion and metastasis of the transitional cell carcinoma of the bladder,so as to provide the new ideas and theory basic for diagnosis,therapy andpredictthe prognosis of transitional cell carcinoma of the bladder. Materials and Methods:By adopting immunohistochemisty SP method, the expressions of CD34 protein and CD105 protein in tissue of 6 papilloma of bladder, 12 cases of grade I of and 18 cases of grade Ⅱ-Ⅲ of (10 cases accompany with the lymph node metastasis)were detected.All the datas were analyzed by SPSS11.0 statistical software.the comparison of positive rates uses the Chi-square and Fisher,Statisticallysignificant level was considered as "alpha equals 0.05"Results: 1.The rates of positive expressions of CD34 protein in the tissue of bladder papilloma and grade Ⅰ of transitional cell carcinoma of the bladder and grade Ⅱ-Ⅲ of transitional cell carcinoma of the bladder are respectively 83%, 58%(15/28) and63%. The difference of pairwise comparisons out of the three groups have statistical significance (P < 0.05).2. The rates of positive expressions of CD34 protein in the tissue of non-metastasis and metastasis are respectively 55%(11/20) and 40%(4/10). The difference in the two groups has statistical significance (P < 0.05) .The rates of positive expressions of CD34 protein in the tissue of non-invasion and invasion are respectively 52%(11/19) and 36%(6/17). The difference in the two groups has statistical significance (P < 0.05).3. The rates of positive expressions of CD105 protein in the tissue of bladder papilloma and grade I of transitional cell carcinoma of the bladder and grade II-III of transitional cell carcinoma of the bladder are respectively 16.0%(l/6), 83.0%(10/12) and 90.0%(16/18), The difference of pairwise comparisons out of the three groups have statistical significance (P < 0.05) .4. The rates of positive expressions of CD105 protein in the tissue of non-metastasis and metastasis are respectively 82.0%(10/12) and 88.0%(16/18). The difference in the two groups has statistical significance (P < 0.05) .The rates of positive expressions of CD105 protein in the tissue of non-invasion and invasion are respectively 48.0%(9/19)and 80.0'%(14/17). The difference in the two groups has statistical significance (P< 0.05).5.The rate of positive expressions of CD34 protein in tissue of bladder carcinoma is closely interrelated to the rate of expressions of CD105 (r=0.018, P < 0.05).Conclusions: 1.The immunohistochemisty is first used to detect the MVD in thetumour tissues whose endothelial cells are marked by CD34 and CD105,furthermore,thereserches; are on the relation between CD34,CD105 and the pathogenesis,developmentof the papillom,grade Ⅰ-Ⅲ of transitional cell carcinoma of the bladder.2.The rar.es of positive expression of CD 105 and MVD are very low in the tissues ofpapillom of bladder, but, in proper order, it obviously increases in tissue of grade Ⅰ, Ⅱand Ⅲ of transitional cell carcinoma of the bladder. It is pointed out that CD105 maybetake part in the pathogenesis and development of transitional cell carcinoma of thebladder. CD 105 are expected to be the biological signs of illness state examination andinitial diagnosis.3. The powerful positive expressions of CD 105 obviously concern the invasion oftransitional cell carcinoma of the bladder and lymph node metastasis. It is pointed outthat CD105 can be regarded as the index of metastasis antcipation of transitional cellcarcinoma of the bladder and can instruct the chemotherapy and radiotherapy afteroperation4.Detecting CD34 and CD105 simultaneously testify that they not only played apositive role to each other in course of pathogenesis, development, invasion ,lymphnode metastasis of transitional cell carcinoma of the bladder.but also they are helpful todicuss the pathogenesis, development, invasion ,lymph node metastasis and predict theprognosis of transitional cell carcinoma of the bladder.5.In signing the MVD of transitional cell carcinoma of the bladder,CD105 is moreparticular than CD34.
