Objective: To investigate the relationship betweenoxidative stress and heat shock protein-60 on the thegenesis , persistence of atrial fibrillation(AF) ,by testingthe levels of C-reactive protein (CRP), SuperoxideDismutase(SOD), Malondialdehyde(MDA) in plasma, andexpression of heat shock protein-60 (HSP60) in sample ofatrium.Methods: Selected 60 patients with atrialfibrillation as AF group, and 60 patients with sinus rhythmas sinus rhythm group. Each group was composed of 30cases with rheumatic heart disease and 30 cases with non-rheumatic heart disease as subgroup (subgroup rheumaticheart disease and subgroup non-rheumatic heart disease).The levels of CRP, MDA ,SOD were tested to each patient.22 cases in AF group and 20 cases in sinus rhythm groupwere selected as subgroup of surgery who sufferedrheumatic heart disease. Atrial samples were excised anddetermined the expression of HSP-60 byimmunohistochemistry.Result: ⑴Levels of plasma MDA and SOD inAF group are increased in significantly comparing to sinusrhythm group. (8.46 ±2.13 nmol/ml vs 6.41 ±2.05nmol/ml; 69.38±12.76 U/ml vs 59.67±14.14 U/ml). ⑵Levels of plasma CRP in AF group are increased insignificantly comparing to sinus rhythm group ( 20.72±17.92 mg/L vs 6.80 ±6.26 mg/L). ⑶In subgrouprheumatic heart disease of AF, levels of plasma MDA, SOD,CRP are close without significantly comparing to subgroupnon-rheumatic heart disease. (8.27±2.11 nmol/ml vs 8.66±2.16 nmol/ml;69.85±10.92 U/ml vs 68.91±14.55 U/ml;21.65±18.34 mg/L vs 19.78±17.75 mg/L). ⑷Observedthe atrial biopsy samples by histology microscopy,hypertrophy of the atrial myocytes and patchy fibrosis insubgroup surgery of AF were more evident than insubgroup surgery of sinus rhythm. ⑸The positive rate ofHSP-60 in subgroup surgery of AF is increased insignificantly comparing to subgroup surgery of sinusrhythm. ⑹There was a negative correlation betweengray degree, which indicate the expression of HSP-60, andlevels of SOD. Coefficient of product-moment correlationis negative 0.54 in significant.Conclusion: Patients with atrial fibrillation suffer acondition of oxidative stress. The increased expression ofHSP-60 may represent a protective mechanism.
