Objective To study the inhibitory effect of dexamethasone(DX) combined with mitomycin C (MMC)by intracapsular infusion method on rabbit lens epithelial cells and its intraocular toxicity.Methods Sixteen rabbits were divided into four groups.Sodium hyaluronate was injected into the anterior chamber to protect epithelium cell of cornea.0.2 ml various drugs:NS(group A), DX( group B,4mg/ml , 0.08mg),combination of DX and MMC(group C, DX 0.8mg,MMC 0.02mg) ,MMC(group D,0.1mg/ml,0.02mg) was injected into intracapsular for hydrodissection , allowed to remain inside for 3 minutes. Then extracapsular lens extraction(ECLE) was performed with CCC capsulorhexis, investigate inflammatory response after surgery. After 3 months, the PCO was evaluated histologically. The expression of bcl-2 and PCNA were detected by immunohistochemical streptavidin-peroxidase(SP).Results The dispearing time of conjunctiva congestion and aqueous humor inflammatory was significantly difference between group B, C and group A, D(P<0.01).The starting time of PCO in every experiment group was later than the control group(P<0.01). Both dexamethasone and mitomycin C , combination of them were effective in preventing PCO compared with the control (P <0.01 — 0.05). Dexamethasone had a weaker effect than the other drugs, it only delayed the time of PCO appearing.On the other hand dexamethasone can reducing postoperative inflammation after ECLE. Combination of DX and MMC significantly prevented the development of PCO in rabbit eyes,and influence the blood-aqueous barrier slightly.
