Experimental Studying The Effect Of Chinese Herbal Medicine BBYH On Melanocyte Metabolism

Objective: to study the effects of Bai Ban Yi Hao (BBYH) on proliferation, activity of tyrosinase and melonogenesis in B16 murine melanoma cells in vitro and vivo for explaining the relationship between BBYH and melanocyte metabolism , describing these mechanisms in the treatment of vitiligo.Methods: B16 murine melanoma cells were cultivated in vitro with five concentrations medicines. The blank group is DMEM and the control group is only melanoma cells. The cytological, enzymological and serum pharmacological methods were used to study the effects of BBYH on proliferation, activity of tyrosinase and melonogenesis in B16 murine melanoma cells. MTT was used to detect the proliferation of melanoma cells, activity of tyrosinase was measured by enzymological method and melanin was detected by its absorption at 405nm.Results:1. BBYH can proliferate B16 murine melanoma cells. Five test groups were significantly different from the control group (P<0.01). Four groups (5~20mg/ml) showed the positive correlation. Whereas when the concentration over 25mg/ml, the toxicity effect of cells showed that values of OD were lower than 20mg/ml group.2. BBYH can enhance activity of tyrosinase.
