Invasion and metastasis are two important characters of malignant tumor,in two process, the PTEN and VEGF play an important role. Objective To investigate the expression of PTEN and vascular endothelial growth factor (VEGF) in renal cell carcinoma(RCC),and the relationship between their expression. Methods Formalin-fixed paraffin embedded tissues from 48 cases of RCC and 18 cases of normal renal tissues which were closely adjacent to carcinoma and 9 cases of normal renal tissues were evaluated for PTEN protein expression by immunohistochemical S - P method .The results were compared with histological grade and clinical stage.VEGF was also detected in 48 cases of RCC and 18 cases of normal renal tissues which were closely adjacent to carcinoma and 9cases of normal renal tissues and its correlation with PTEN was concerned. Results The positive incidence of expression PTEN protein in RCC was 45.8% which was prominently lower than those in the adjacent tissues of RCC (88.9%) and normal renal tissues (100.00%)(P<0.05). The expression of PTEN was negatively correlated with histological grade ,clinical stage and prognosis. The positive incidence of expression VEGF protein in RCC was 68.8% which was prominently higher than those in the adjacent tissues of RCC (27.8%) and normal renal tissues (22.2%)(P<0.05). The expression of VEGF was notable correlated with histological grade and clinical stage. There is trend that PTEN negatively correlated with VEGF. Conclusion Loss or decreased expression of PTEN is markedly correlated with clinical stage and histological grade, and PTEN negatively correlated with the expression of VEGF. Combined detection of PTEN and VEGF may be helpful to evaluate prognosis and infiltrative capability of RCC.
