Objective:To explore the influence of QiLeng decoction on nerve cell apoptosis after focal cerebral ischemic reperfusion in rats ,and to provide the scientific foundation for benefit vital energy (BVE), nourishing yin(NY),activating blood circulation(ABC) on treating ischemic stroke. Methods:The rats were randomly divided into four groups including sham-operation group,model group, BuYangHuanWu decoction group and QiLeng decoction group. The model of focal cerebral ischemia reperfusion injury were was made by using thread embolizing and fastening of the TCA in rats. TUNEL was employed to measure the cell apoptosis and Caspase-3 and Spectrin protein were observed by immunohistochemical method at different time point after cerebral ischemia reperfusion. The cerebral cell morpathology was examined by HE. Results:(1) The cerebral structure in sham-operation group was normal, while that in the modle group showed ischemic.Contrasting with model group, BuYangHuanWu decoction group and QiLeng decoction group could improve the ischemic pathological changes of brain in rats much better at the end of 24th hour after reperfusion ,and the effect of QiLeng decoction group is superior to that of BuYangHuanWu decoction group.(2) The apoptosis in model group were significant more increased than those in sham-operation group(P<0.01). The apoptosis in BuYangHuanWu decoction group and QiLeng decoction group were all reduced significantly compared with the model group (p<0.01).(3)The expression of Caspase-3 protein in model group was significantly higher than that in sham-operation group(P<0.01), and the expression of Caspase-3 in BuYangHuanWu decoction group and QiLeng decoction group were all reduced significantly compared with the model group (P<0.05).(4) The expression of Spectrin protein in model group was significantly less than that in sham-operation group(P<0.01), and the expression of Caspase-3 in BuYangHuanWu decoction group and QiLeng decoction group were all increased significantly compared with the model group (P<0.05).(5) Contrasting with BuYangHuanWu decoction group, QiLeng decoction group could save the apotosis ,decrease the expression of Caspase-3 protein andincrease the expression of the Spectrin protein. Conclusion:QiLeng decoction play the neuron protective effects by restraining the expression of the Caspase-3, and decreasing the decomposing of Spectrin, reducing the neuron apoptosis. It may be one of the mechanism of benefit vital energy (BVE), nourishing yin(NY),activating blood circulation(ABC) treating ischemic stroke.
