Objective:To investigate the mechanisms of Qileng Decoction and the therapeutic method of Benifiting Vital Qi (BVQ), Nourishing Yin(NY) and Activating Blood Circulation (ABC) against focal Cerebral ischemia- reperfusion injury ,so as to provide laboratory evidence for clinical use.Methods:The rats were randomly divided into 5 groups including the native group, the sham-operation group, the MCAO (Middle Cerebral Artery Occlusion) group, the group of Qileng Decoction without nourishing-Yin herbs(the control group) and the Qileng Decoction group(the treatment group), the MCAO model was established with method of internal carotid artery occlusion by suture. Calpain1 and MAP2 were observed with immunohistochemistry method, and apoptosis cells were detected with TUNEL at different reperfusion time of Oh, 2h, 4h, 24h and 48h.Results:There were few Calpain positve or apoptosis cells observed in the native group while MAP2 was extensively expressed. The Calpain positve and apoptosis cells dramatically increased in the MCAO, control and treatment groups after cerebral ischemia-reperfusion, both with a summit at the reperfusion time of 24h and with another smaller summit at 2h for Calpain.
