Objective:To investigate the mechanisms of Qileng Decoction and the therapeutic method of BVQ(Benifiting Vital Qi), Nourishing Yin(NY) and Activating Blood Circulation(ABC) against focal cerebral ischemia-reperfusion injury, so as to provide laboratory evidence for clinical use.Methods:The rats were randomly divided into 5 groups including the native group,the sham-operation group,the MCAO(Middle Cerebral Artery Occlusion) group, the group of Qileng Decoction without nourishing-Yin herbs(the control group) and the Qileng Decoction group(the treatment group).the MCAO model was established with method of internal carotid artery occlusion by suture. Neurography grade were evaluated at6h,1d,2d,3 d and 7d after cerebral ischemia reperfusion, and the pathology changes was observed at 24h reperfusion time.Immunohistochemical method was performed to examine the expression of t-PA and PAI-1 method at different time point after reperfusion.Results:(1)QLD and its decomposed formulas compared with NS control, neurologic scores at different reperfusion time were improved and improved pathology changes were observed at 24h after cerebral ischemia reperfusion. The effect of QLD group is superior to that of QLDWNY group.(2)The exprssion of t-PA and PAI-1 levels and activities in neurons is up-regulated with the treatment of QLD and QLDWNY. The effect of QLD group is superior to that of other group.Conclusion:QiLeng decoction has good effects against cerebral ischemia-reperfusion injury which may be due to its function of inhibiting t-PA and PAI-1, in which treatment method of Nourishing Yin may play an important role.
