【Objective】To detect the expression of myelin protein zero in inner earsand peripheral nerve system; To analysis the differences in the sequences andquantity of myelin protein zero between inner ears and peripheral nervesystem and try to explain the different pathogenicity in these positions; Todetect the distribution and integrate the understand of the real location ofmyelin protein zero in inner ears.【Method】The sequences and quantity of myelin protein zero between innerears and sciatic nerves were analysised by sequence determined andsemi-quantitative RT-PCR ; The location of myelin protein zero in inner earswas detected by immunohistochemistry .【Result】Myelin protein zero is expressed in rats'inner ears and sciaticnerves. In rats'inner ears, it is localled in the auditory nerve and the spiralganglion, while not in the Organ of Corti and basilar membrance. There is nosingnificant difference in the sequences between inner ears and sciaticnerves , while the mRNA in inner ears(R1) and sciatic nerves (R2) are0.5235±0.15684 and0.3333±0.19937 respectively ,and P﹤0.05 by pairedsamples test, thus the expression in inner ears is higher than which in sciaticnerves.【Conclusion】There is no singnificant difference in the sequences between inner ears and peripheral nerve system. The expression of myelin proteinzero in inner ears is higher than which in sciatic nerve. The difference of thepathogenicity might relate to the different expression in these positions. Inrats'inner ears, myelin protein zero is localled in the auditory nerve and thespiral ganglion, which fitly coincide with the potential affection position ofauditory neuropathy. Myelin protein zero might relate to auditoryneuropathy.
