Traditional Chinese Medicine Of Peripheral Neuropathy In Diabetes Rat Sciatic Nerve Cell Apoptosis And Bcl-2, Bax Influence

Purpose: Preliminary discussion on the different extraction liquid herbal for diabetic peripheral neuropathy rat sciatic nerve cell apoptosis and the regulation of gene bcl– 2,bax influence.Method: Rats with intraperitoneal injection STZ to copy diabetes peripheral neuropathy model, will diabetic rats randomly divided into model group, water extraction liquid group, water extraction and alcohol heavy fluid group, ethyl acetate extract group, and to make healthy rats normal control group, during the experiment, water extraction liquid group, water extraction and alcohol heavy fluid group, ethyl acetate extract groups were given corresponding extract, model group and control group given the physiological saline with volume. Eight weeks after separation rat sciatic nerve fibers, all with in situ end labeling method (dUTP TdT - mediated, Nick end labeling, TUNEL) detection sciatic nerve cell apoptosis, immunohistochemistry methods were used detection regulation of gene bcl - 2 and bax protein expression.Result: (1) the blood sugar changes: compared with normal control group, the rats were significant rise in blood sugar, compared with model group, water extraction liquid good blood glucose control group (P < 0.01), and the remaining two group of blood sugar without obvious change; (2) the influence of apoptosis, compared with normal control group, each rat sciatic nerve fibers were significantly increased, the apoptotic cells compared with model group, each rat sciatic nerve fibers were significantly reduce the apoptotic cells (p < 0.05), including among them with ethyl acetate group and water extraction group effect is better; (3) the regulation of gene bcl - 2 ,bax of influence: water extraction liquid group and ethyl acetate extract group significantly reduce the expression of promote apoptosis genes bax (P < 0.05), water extraction liquid group significantly increased the expression of antiapoptotic genes bcl - 2 (P < 0.05).Conclusion: TCM compound can reduce the expression of promote apoptosis genes bax, increase antiapoptotic genes expression of bcl-2, reduce the sciatic nerve cell apoptosis, for the clinical treatment of diabetic neuropathy mention objective basis.
