Objective To investigate the effects of Carthamus tinctorius on infarction volumes,apoptosis of neurons and bcl-2,caspase-3 expressions after acute injury of cerebral ischemia and reperfusion.Methods SD rats were randomly divided into ischemia control group,Carthamus tinctorius large-dose group, middle-dose group low-dose group, Ge gen su group. The Middle cerebral artery of rats was occluded for 2h by inserting an introluminal molofilament, and reperfusion was then instituted for 4h or 22h.The brains were stained with 2,3,5-triphenylte trazolinm chloride for assessment of volume of infarction, and then embedded onto slides with paraffin for morphological assessment and immunohistochemistry wascarried out to investigate the changes in bcl-2 and caspase-3. SD rats were randomly divided into ischemia control group,Carthamus tinctorius large-dose group,middle-dose group,low-dose group,Ge gen su group.The middle cerebral artery of rats was occluded for 2h by inserting an introluminal molofilament and reperfusion was then instituted for 22h.The apoptic rate of each group was measured by flow cytometry.Results All treated groups at different times decreased the volume of infarction (P<0.05) ,while large-dose group and Ge gen su group showed more distinct decrease than other groups (P<0.05).A\\ treated groups at different times increase bcl-2 and decrease caspase-3 expression as well, while large-dose group and Ge gen su group showed more distinct effect than other groups (P<0.05).The evaluations of neurological function of treated groups were lower than ischemia control group,and those had statistic meaning(P<0.05),while large-dose group and Ge gen su group showed more distinct effect than other groups.(P<0.05).The apoptotic rates of treated groups were lower than ischemia control group,and those also had statistic meaning(P<0.05),while large-dose group and Ge gen su group showed more distinct decrease than other groups (P<0.05).Conclusions Carthamus tinctorius can reduce the volume of cerebral infarction, and increase bcl-2 and decrease caspase-3 expression.Carthamus tinctorius can reduce the apotosis of cerebral cells after ischemia and reprfusion.Carthamus tinctorius can reduce the synptom of ischemia and have the effect of cerebral protection.
