OBJECTIVE1. To investigate the levels of the serum soluble endothelial growth factor receptor-1 and placental growth factor in women with preeclampsia and compare the difference between the mild preeclampsia, sever preeclampsia and eclampsia。 Foremore to analyse the association of the levels of the serum soluble vascular growth factor receptor-1 and placental growth factor and preeclampsia.2. To detect the expression of the membrane-bound VEGFR-1 protein in - the placenta tissue of the women with preeclampsia and compare thedifference between the mild preeclampsia, sever preeclampsia and eclampsia。 Then to analyse the association of the expression of the membrane-bound endothelial growth factor receptor-1 and preeclampsia。METHODS AND RESULTS10 women with eclampsia (EP), 10 women with mild preeclampsia (MP), 10 women with sever preeclampsia (SP) and 10 women without preeclampsia (NC) were selected as study groups and control group。 1. The change and significance of serum levels of soluble vascular growthfactor receptor-1 and placental growth factor in women with preeclampsia: The serum levels of sVEGFR-1 and PLGF are detected by the quantitivesandwich enzyme immunoassay technique。1) The results showed that the serum sVEGFR-1 levels of eachpreeclampsia groups were relatively higher than those of control group(P<0.05) and that there were significant differences between the mildpreeclampsia, sever preeclampsia and eclampsia。(2) The serum concentration of PLGF of preeclampsia groups was relatively lower higher than those of control group (P<0.05) and there were significant differences between the mild preeclampsia, sever preeclampsia and eclampsia (P <0.05)。(3) The ratio of the sVEGFR-1 concentration and the PLGF levels (sVEGFR-1/PLGF) in serum of preeclampsia groups is relatively higherthan that of control group (p<0.05) and there were significant differencesbetween the mild preeclampsia, sever preeclampsia and eclampsia (P <0.05)。2. Quantitive and local study of membrane-bound VEGFR-1 in theplacenta tissue of preeclampsia:Immunohistochemistry and quantitive analyse were used to detect membrane-bound VEGFR-1 contents and distribution in the placenta tissue。(1) In placentas positive staining was detected predominantly in the membranes of villus synsytiotrophoblast cells, the membranes of extravillus trophoblast cells, the membranes of villus endothelial cells. In fetal membranes positive staining was detected predominantly in amnionic endothelial cells.(2) The contents of membrane-bound VEGFR-1 in placenta ofpreeclampsia groups were relatively lower than that of control group. There were significant differences of membrane-bound VEGFR-1 contents between the mild preeclampsia, sever preeclampsia and eclampsia (P <0.05),(3) The ratio of the sVEGFR-1 concentration in serum and the membrane-bound VEGFR-1 contents in placenta of preeclampsia groups (sVEGFR-1/ membrane-bound VEGFR-1) is relatively higher than that of control group (f<0.05) and there were significant differences between the mild preeclampsia, sever preeclampsia and eclampsia (P <0.05) 。CONCLUSION1. The serum PLGF levels of each preeclampsia groups were relatively lower than those of control group . There may be an association between preeclampsia and PLGF。2. The serum sVEGFR-1 levels of preeclampsia groups were relatively higher than those of control group. VEGFR-1 may be related to preeclampsia.3. The contents of membrane-bound VEGFR-1 in placenta of preeclampsia groups were relatively lower than that of control group 。 Membrane-bound VEGFR-1 may relate to preeclampsia。4. The ratio of the sVEGFR-1 concentration and the PLGF levels(sVEGFR-1/PLGF) in serum of preeclampsia groups is relatively higher than that of control group. The ratio of the sVEGFR-1 concentration in serum and the membrane-bound VEGFR-1 contents in placenta of preeclampsia groups(sVEGFR-1/membrane-boundVEGFR-1)is relatively higher than that of control group。 Preeclampsia may congtribute to VEGFR-1/PLGF and sVEGFR-1/membrane-bound VEGFR-1.
