Studies Of The Anti-tumor Activity Constituents On Clematis Manshurica Rupr

Clematis manshurica Rupr is from clematis of buttercup. It was reported that Saponins of Clematis manshurica Rupr had significant anti-tumor effect and it had anti-tumor effect on mice bearing sacocarcinoma S₁₈₀. But studying on anti-tumor effect components of grease part of Clematis manshurica Rupr were accordingly not reported. In order to. search for and make sure its anti-tumor effectives components, the ethl acetate fraction of the Clematis manshurica Rupr were studied in this experiment. The report elucidated their pharmacological effect and chemical constituents.The ethyl acetate extract fraction of Clematis manshurica Rupr were firstly used for measuring the anti-tumor effect on mice bearing sacocarcinoma S₁₈₀. Then we have isolated the compounds from the ethyl acetate fraction, and its anti-tumor activity were measured on Hela cell.The results showed: (I) The ethyl acetate fraction have been showed anti-tumor activity on sacocarcinoma S₁₈₀. (II) 7 compounds were isolated from the ethyl acetate fraction by methods of repeated silica gel column chromatography. Among them , 7 components chemical structures were identified by the means of physiochemical properties, spectrography and comparing with standard compounds. Their chemacal structure were shown below respectively: (1) β -sitosterol. I, (2)Oleanolic acid, (3)coumarin, (4)Myristic Acid, (5)Palmy Acid, (6)Quercitin, (7)Oleanolic - O-β-D-glucoside. (III) The results of anti-tumor test showed that (2), (6) had anti-tumor to Hela cell.Above all: Ethl acetate fraction of Clematis manshurica Rupr was shown anti-tumor activity, compound (7)was isolated from Clematis manshurica Rupr for the first time, compound (2), (6) also shown anti-tumor activity, the other active components remain to be studied in the future.
