Objective:To study the chemical constituents and their anti-inflammatory activities from n-BuOH extract of the roots of Clematis chinensis Rupr.Methods:The n-BuOH faction of the 70%EtOH extract of the roots of C.chinenis was purified by using normal-phase and reversed-phase silica gel,medium and high performance liquid chromatography.The structures of the isolated compounds were elucidated by NMR spectroscopy.The isolated compounds were evaluated for their inhibitory effects against the pro-inflammatory mediators nitric oxide(NO),tumour necrosis factor-a(TNF-a)and Interleukin-6(IL-6)in lipopolysaccharide(LPS)-stimulated mouse macrophages cells(RAW 264.7)by Griess and ELISA methods.Results:Twelve compounds 1-12 were isolated from the n-BuOH extract of the roots of C.chinenis.The structures of compounds were identified as oleuropein(1),liguseroside(2),oleoside dimethyl ester(3),syringing(4),coniferin(5),sinenside ?(6),aglycones olivil(7),hyuganoside ?b(8),3,4-dihydroxyphenyl(9),3-(2-aminoethyl)catechol(10),?-D-fructof uranose(11)and 2-(4-hydroxy-3-methoxyphenyl)ethanol l-0-[?-D-apiofuranosyl-(1?6)-?-D-glucopyranoside(12)by comparing their spectral data with those of published in literatures.Among of them,compounds 3,5,6,7 and 8 were obtained from C.chinensis for the first time.The anti-inflammatory effects of the isolated compounds 1-12 were evaluated in terms of inhibition of production of NO,TNF-? and IL-6 in LPS-stimulated RAW 264.7 cells.Compounds 1,2 and 9 were found to possess better inhibitory activity than the other compounds tested.
