objective: To study the expression of NF-κBp65 and IκBα in nasal polyps(NP),uncinate process mucosa of chronic rhinosinusitis and inferior turbinates(IT);To observe the effects of systemic corticosteroids on the expression of NF- κ Bp65 and IκBα in nasal polyps.Method:The specimens from 30 nasal polyps,20 uncinate process mucosa of chronic rhinosinusitis and 20 inferior turbinates were studied by SP immunohistochemical methods for NF- κ Bp65 and IκBα. The SP immunohistochemical methods were applied to explore the expression of NF-κBp65 and IκBα in nasal polyps before and after corticosteroids therapy.The optical density of positive cells in mucosal epithelia,glandular epithelia,vascular endothelia, inflammatory cells was measured by using HPIAS-2000 image-conduct system.Result:(1)The expression of NF-κBp65 was positive in the cytoplasm and some unclear of the mucosal epithelia,the inflammatory cells, the glandnar epithelia and the vascular endothelia in nasal polyps.(2)The expression of IicBα was positive in the cytoplasm of the mucosal epithelia,the inflammatory cells ,the glandnar epithelia and the vascular endothelia in nasal polyps.(3)The expression of NF-κBp65 was significantly higher in the mucosal epithelia , inflammatory cells , glandular epithelia and vascular endothelia of nasal polyps than that in uncinate process mucosa of chronic rhinosinusitis and inferior turbinates(P<0.05, P<0.01). The expression of NF-κBp65 was significantly higher in the mucosal epithelia , inflammatory cells , glandular epithelia and vascular endothelia of uncinate process chronic rhinosinusitis than that in inferior turbinates(P<0.05).(4)The expression of IκBα was significantly higher in the mucosal epithelia , inflammatory cells , glandular epithelia and vascular endothelia of nasal polyps than that inuncinate process mucosa of chronic rhinosinusitis and inferior turbinates(P<0.05, PO.01). The expression oflKBa was significantly higher in the mucosal epithelia , inflammatory cells , glandular epithelia and vascular endothelia of uncinate process chronic rhinosinusitis than that in inferior turbinates(P<0.05).?The expression of NF-icBp65 was significantly reduced and the expression of IkBcx was significantly increased in the mucosal epithelia and inflammatory cells of nasal polyps after treated with corticosteroids compared with pre-treated.?Compared with pre-treated, the expression of NF-KBp65 and IkBoi in glandular epithelia, vascular endothelia of nasal polyps has no statistically difference (P>0.05).Conclusion:CDNF-KBp65 and IicBa might play a key role in the pathogenesis of nasal polyps.?NF-icBp65 and IicBa might play a key role in the pathogenesis of chronic rhinosinusitis.?Arosing the expression of IkBci in mucosal epithelia and inflammatory cells of nasal polyps inhibiting the expression of NF-icBp65 in those cells may be one of mechanisms of systemic corticosteroids therapy on the nasal polyps.
