| OBJECTIVESUlcerative colitis (UC), which is also called non-specificity ulcerative colitis ,is an agnogenically inflammatory disease of rectum and colon . The lesion mainly occurs stratum mucosum and stratum submucosum. Diarrhea ,stomachache and blennoid bloody purulent sstool are the clinical manifestations. Patients' conditions are different but most of those attack recurrently on the course of their chronic diseases.The etiopathogenisis and nosogenesis are unclear now . Most researchers think they are related with infection, immunologic abnormality, heredity and spirit and so on . Varied predisposing factors provoke intestinal mucosa of the susceptibles and radical immunoinflammatory reaction occur .Recently the intimate relationship between functional abnormality of organism immunity and nosogenesis of UC. The evidences are as follow :the patients with UC usually have extraintestinal complications which are caused by functional abnormality of organism immunity, such as multiple arteritis, Sjogren Syndrome, thyroiditis, diabetes mellitus,autoimmune hemolytic anemia ,erythema nodosum , arthritis, uveitis, iritis,systemic lupus erythamatosus et al .The extent of these diseases are tightly related with the lesion degree and extent of UC .With the relieve of UC, the extraintestinal complications can cure spontaneously .At the same time, anti-colonic -epithelial cell antibody can be detived in serums of some patients and theapplication of adrenocortical hormone and immunosuppressants can relief the UC. All of those hint UC is related with immunity. Mucin (MUC) is a glycosylated protein with high molecular weight ( ) 200kD ) excreted by the epithelial cells of gastrointestinal ducts and ovary. It can lubricate and protect normal mucosas .Once the immunal function of these epithelial cells is disturbed, the quality and quantity of Mucin are changed .The protective effect of mucin in intestinal immunity are thinked more and more highly .Mucins of human colon are encoded by a family with 9 genes, which is MUC1-MUC8 , including MUC5AC and MUC5B.The names of mucins are same with the according genes .The most important one of mucins in normal colon is MUC2.The genic production of MUC2 is the first secreting mucin which sequence is known clearly .Similar with other 8 genic productions ,its sequence includes highly irregular amino acids repeated sequence .By our study is mainly order to discuss the effect of the expression of MUC2 in etiopathogenisis of ulcerative colitis and supply the experimental evidences for the researches on nosogenesis of ulcerative colitis. MATERIALS AND METHODS1. PatientsWe investigated the activation of MUC2 in biopsies from 42 patients aged from 16 to 71 with an acute episode of ulcerative colitis from Mar 2003 to Apr 2005, in which have 28 male cases and 14 female cases.30 cases of normal controls were investigated too.2. Reagents: The mouse monoclonal antibody, which selectively binds to the NLS sequence of the activated MUC2 subunit, was purchased from santa Cruz Biotechnology.3. Immunohistochemical staining of tissue sections: Sections 4pm thick were deparaffinized in xylene and rehydrated through degraded concentrations of ethanol .The expression of MUC2 was examined in 60 cases of paraffin-embedded tissues by immunohistochemical methods, which were from endoscopic biopsies of 42 cases active UC and 30 cases normal controls.4. Statistical Methods : All of the data were recorded and set up data bank which was performed statistical analysis by SPSS 10.0 software. The level of significance was p<0.05. Databank was analyzed by t test and signed rank test. RUSULTS1. MUC2 were expressed in all the normal colitis tissues ,mainly in epithelial cells and endothelial cells in variant degrees .2. MUC2 were localized in the cytoplasm of cells.3. MUC2 were all expressed in normal controls. There are significant differences between UC patients and normal controls (83.1 + 3.8 vs 100 ± 1.1) PO.01.4. The expression of MUC2 was partly related to the activity of ulcerative colitis, such as severity, endoscopic classification, pathologic classification and disease extent. The expression of MUC2 as significant difference between mild and severe cases (92.1 ±3.2vs73.5±2.1, PO.05 ). The expression of MUC2 has significant difference between endoscopic class I and class II (91.3±3.5vs83.7±2.4, p<0.05), class I and III (91.3 ± 3.5vs74.3 ± 1.9, PO.01). Significant difference was also between pathologic class I and II (92.4±4.0vs85.9±2.3, PO.05), class I and III (92.4 + 4.0vs73.7±2.1,P<0.01).CONCLUSIONS1. The induction of MUC2 participates in the occurrence and development of UC. It may play an important role in the pathogenesis of ulcerative colitis.2. MUC2 activation may be related with the severity and activity of UC.3. MUC2 were localized in the in the cytoplasm.4. MUC2 were expressed in normal controls .On the contrary, MUC2 were little expressed .It demonstrates that the decrease of MUC2 activation related to inflammation. |
