Effect Of Shuanghongqinyu Decoction On The Content Of IL-23and MUC2 Expression In Rats With Ulcerative Colitis

Objective: This reasearch by observing the influence of Shuanghongqinyu decoction to ulcerative colitis rats?general state,disease activity index(DAI)score,colonic mucosa pathological changes,serum level of interleukin 23(IL-23)and colonic mucosal tissue mucin2(MUC2)expression,discussing the possible mechanism of Shuanghongqinyu decoction to the ulcerative colitis.Methods: Sixty healthy male Wistar rats(weighting at 250-300g),after adaptive feed for one week,were randomly divided into six groups: the control check(CK),model group(MG),mesalamine group(MSG),Shuanghongqinyu decoction high dose group(HDG),moderate dose group(MDG)and low dose group(LDG).Each group contained ten rats.Except the control group,other groups rats were administered with 2,4,6-trinitrobenzene sulfonic acid(TNBS)to induce ulcerative colitis model.Four days after inducing model,the CK and MG rats were fed as uaual,the remaining four groups rats were given mesalamine and different concentration of Shuanghongqinyu decoction respectively.All of them were treated for two consecutive weeks.During the period of treatment,we observed rats mental state,body weight,stool,food,hair color and so on,and counted each group DAI score.After treatment,all rats fasted for twenty four hours,were killed to collect blood samples to measure serum IL-23 levels by ELISA,and colonic mucosal tissues were taken to detect the expression of MUC2 by immunohistochemistry method.Results:1 general state and DAI scoreGeneral state: NK rats in good spirit,smooth hair,food intake as uaual,flexible activities and normal stool.The rest five groups rats in depression,lassitude,slow in reacting,significant reduction of food intake,hair disorder,back arched and loose stools.MG rats symptoms gradually worse with increase of experiment days.The four treatment groups rats had an improved in general condition,food intake,stool after drug therapy,of which HDG close to CK,MSG and MDG showed no significant difference but better than LDG.DAI score: The results showed that CK(0.000±0.000)was significant lower than other five groups(P<0.05);the four treatment groups scores were lower than MG(2.485±0.243),statistically significant difference(P<0.05);the HDG(0.406±0.282)score was lower than MSG(0.899±0.298),MDG(0.901±0.357),LDG(1.332±0.267),statistically significant difference(P<0.05);the score of LDG(1.332±0.267)was higher than MSG(0.899±0.298),MDG(0.901±0.357),statistically significant difference(P<0.05);while MSG(0.899±0.298)and MDG(0.901±0.357)showed no significant difference(P>0.05).2 colonic mucosal tissue pathological observationNaked eye: MG rats colonic mucosal tissue with most injuries,visually worst;HDG close to CK;MSG and MDG better than LDG;LDG slightly better than MG.Pathological observation: CK mucosal epithelium smooth,normal gland with clear structure;MG mucosal with obvious hyperemia,edema,necrosis and ulcerations;HDG nearly integrity,close to CK;MDG and MSG rat colonic tissue inferior than LDG and better than LDG.3 the level of IL-23 and the content of MUC2IL-23: The level of IL-23 in CK(21.07±0.87)was significant lower than others(P<0.05);the level of four treatment groups were lower than MG(31.30±1.65),the difference with statistically significant(P<0.05);HDG(22.46±0.60)was significant decreased than that of MSG(24.85±1.17),MDG(25.42±1.18)and LDG(27.82±0.81)(P<0.05);LDG(27.82±0.81)was higher than MSG(24.85±1.17)and MDG(25.42±1.18),statistically significant difference(P<0.05);while MSG(24.85±1.17)and MDG(25.42±1.18)with no significant difference(P>0.05).MUC2: The determination of MUC2 expression in CK(0.415±0.032)was higher than other groups,the difference was statistically significant(P<0.05);four treatment groups were significant increased than MG(0.062±0.006)(P<0.05);HDG(0.298±0.040)was higher than that of MSG(0.146±0.055),MDG(0.129±0.034)and LDG(0.096±0.029),statistically significant difference(P<0.05);LDG(0.096±0.029)was sifinificant decreased than MSG(0.146±0.055)and MDG(0.129±0.034)(P<0.05);while the difference of MSG(0.146±0.055)and MDG(0.129±0.034)was no significant(P>0.05).Conclusions:1 Shuanghongqinyu decoction can effectively improve the ulcerative colitis rats general state,reduce DAI score,relieve colonic mucosal pathological damage and show a certain dose-effect relationship.The result of HDG is the best.2 The possible mechanism of Shuanghongqinyu decoction on the treatment of UC by down-regulation of serum IL-23 and up-regulation of MUC2 expression to adjust colonic immunoreaction and promote the repair of damaged mucasa.
